Objective Research on the electrochemical behaviors of mouse fibroblasts(3T3), human breast cancer cells(MCF-7) and melanoma cells(B16). Study on the changing of electrochemical signals of three cells at different cell secretion time. Established electrochemical methods to explore the difference between normal cells and tumor cells. Xanthine oxidase was added in the cell lysates of three cells, in order to establishment the method to detect the content of adenine(A), guanine(G), xanthine(X) and hypoxanthine(H) by electrochemical method.Methods In this paper, 3T3, MCF-7 and B16 cells are experimental cell models. Studied the electrochemical behaviors of 3T3, MCF-7 and B16 cells by cyclic voltammetry. The electrochemical signals of 3T3, MCF-7, B16 cells have been assigned by high performance liquid chromatography. Detecting the changes of electrochemical signals of three cells in different secretory times and the content of adenine, guanine, xanthine and hypoxanthine in three cells by electrochemical method.Results In the experiment of cells secrete, when 3T3 cells secreted in 60 minutes, the electrochemical signals, peakⅠ and peakⅡ, were maximum. When MCF-7 cells secreted in 40 minutes, the electrochemical signal, peakⅠ and peakⅡwere maximum. When B16 cells secreted in 50 minutes, the electrochemical signal, peakⅠ and peakⅡ, were maximum. The result showed that the secretory capacity of tumor cells were higher than normal cells. In the experiment of quantitative analysis of the content of four purines in 3T3, MCF-7 and B16 cells, after the addition of xanthine oxidase, electrochemical signal of three cell, peakⅠand peakⅡ, were reduced, and produced uric acid peak. By calculated subtraction the amount of reduction and original amount of peakⅠ and peakⅡ, acquired three cell content of four purine.Conclusion The present study investigated the electrochemical signals of three cells in different time cells secrete by electrochemical method. Established electrochemical method to explore the difference between normal cells and tumor cells. Adding xanthine oxidase in cell lysates of three cells, build the way to detect the content of four purines in cells by electrochemical method.
