Objectives To study the expression of NRP-1 and NRP-2 in gastric cancer tissues and the relationship between the expression of NRP-1 and NRP-2 and clinical pathological features in gastric cancer, and explore their value in judging the prognosis of gastric cancer.Methods We analyzed the clinical pathologic data and prognosis of 168 cases with gastric cancer. Detect the expression of NRP-1 and NRP-2 in gastric cancer and normal tissues with immunohistochemical analysis. We investigated the relationship between the expression of NRP-1、NRP-2 and the clinical pathological features in gastric cancer with Pearson Chi-Square test. Kaplan-Meier method was used to analyse the relationship between expression of NRP-1 and NRP-2 level and prognosis of gastric cancer and Logrank test was used to analyze the factors of survival time for any significant differences.Cox regression model was used for multivariate analysis, then we evaluated if NRP-1 and NRP-2 could be used as indexes for prognosis in gastric cancer or not.Results 1 In 168 patients, high expression of NRP-1 positive rate of gastric cancer tissues(66.7%,112 cases) was higher than relatively normal tissue(8.33%,14 cases)(P<0.05). high expression of NRP-2 positive rate of gastric cancer tissues(70.2%,118cases) was higher than relatively normal tissue(10.7%,18 cases)(P<0.05). 2.The expression of NRP-1 was correlated with the tumor size,differentiation degree, infiltrative degree, lymph node metastasis and TNM stage(P<0.05). The expression of NRP-2was correlated with differentiation degree, infiltrative degree, lymph node metastasis and TNM stage(P<0.05). 3 Median survival time with high NRP-1 expression was 27 months and median survival time with low NRP-1 expression was 64 months( χ2=49.326, P=0.000,log-rank test); Median survival time with high NRP-2 expression was 29 months and median survival time with low NRP-1 expression was 50 months( χ2 =13.008, P=0.000, log-rank test). 4 Median survival time with co-high expressionof NRP-1 and NRP-2 was 27 months; median survival time with high expression of NRP-1/NRP-2 was 35months; median survival time with high expression of only NRP-2 was 43 months; median survival time with co-low expression of NRP-1and NRP-2 was 63 months and and the difference was statistically significant(χ2=60.709,P=0.000, log-rank test). 5 The expression of NRP-1 was an risk factor for gastric cancer(95%CI 1.006~1.989,P=0.046). The expression of NRP-2 was an risk factor for gastric cancer(95%CI 1.006~2.941,P=0.048).Conclusion 1 NRP-1 and NRP-2 played important roles in the incidence and development process of gastric cancer and the expression of NRP-1 and NRP-2 was closely related with the malignant biological behavior of gastric cancer. 2 The high expression of NRP-1 and NRP-2 predicted poor prognosis. 3 NRP-1 and NRP-2 maybe expected to be regarded as index to prognosis.
