Objective Wnt protein is a family of glycoprotein abundant in cysteine. Wnt signaling pathway include classical Wnt/β-catenin pathway and non- classical Wnt/PCP、Wnt/Ca2+pathway. Classical pathway function to promote cell proliferation and non-classical pathway has antag effect on classical pathway. Wnt-5a protein, a member of Wnt family,plays central role in the growth and migration of vascular endothelial cell and fibroblast.Aberrant activation of Wnt-5a gene contributes to the tumorigenesis. Wnt-5a is a dual function protein in various tumors due to complex signaling pathway. High expression of Wnt-5a in gastric cancer and non small cell lung cancer can induce tumor metastasis and low expression of Wnt-5a in endometrial cancer and melanoma can inhibit tumor metastasis. The mechanism about Wnt-5a gene in prostate cancer is unclear. Hence, the main goal of this study is to detect the important role of the Wnt-5a in progression of prostate cancer and discuss the correlation of Wnt-5a with prostate cancer development and clinical significance.Method Eighty patients with prostate cancer who underwent surgery at urinary department of Tangshan People’s Hospital and the Second Hospital of Tianjin Medical University from september, 2011 to september, 2014 were entered in this study and 80 patients with hyperplasia of prostate gland were chosen as control. All specimens were confirmed by the pathologist and clinical data were intact. Wnt-5a m RNA level in tumor and non-tumor tissues was measured by real-time quantitative PCR and immunohistochemistry stainning was preformed to cetect Wnt protein expressiion in tumor and non-tumor tissues.The correlation of Wnt-5a expression with clinical parameters was statistically analysed to evaluate the Wnt-5a contribution in development process of prostate cance.Results The relative amount of Wnt-5a m RNA in prostate cancer tissues and hyperplasia tissues of prostate gland was(2.08±0.22) and(0.28±0.02) respectively. The statistics difference was found between two groups(P<0.05). Compared with the hyperplasia tissues, there is noted increase in positive rate of Wnt-5a expression(75.00% vs 27.50%,P<0.05). Wnt-5a protein expression is located in cytoplasm and the positive rate of expression is higher in prostate cancer tissues than in hyperplasia tissues(96.25% vs62.5%, P<0.05). Our results indicat that expression of Wnt-5a m RNA and protein correlate with preoperative PSA concentration in plasma, Gleason score, lymph node metastasis, vascular invasion and perineural invasion(P<0.05)but do not with tha age,tumor stage, seminal vesicle invasion and surgical margin(P>0.05). The Wnt-5a expression increase in the specimens with high PSA concentration, high Gleason score,lymph node metastasis, vascular invasion and perineural invasion, which means that high expression of Wnt5 a gene may be involved in invasion and metastasis.Conclusion Aberrant activation of Wnt-5a may play an improtant role in process of prostate carcinogenesis and development. High expression of Wnt-5a contribute to prostate cancer metastasis, which indicates that Wnt-5a is a oncogene in prostate cancer development. Wnt-5a may be a molecular indicator for diagnosis, monitor and prognosis evaluation in clinic.
