Life Sciences have been developed rapidly over the last decades. Cell line is an important material in life science research.Reliable data depends on the high quality of the cell lines. Providing high-quality cell lines for research is the purpose of national cell bank. Mycoplasmal contamination of cell cultures is a common occurrence that is capable of altering of normal cell structure and function. Mycoplasmas will inhibit cell metabolism and growth, alter nucleic acid synthesis, affect cell antigenicity, induce chromosomal alterations, interfere with virus replication, and mimic viral action. So it is very necessary to establish methods for detecting and removing Mycoplasmas in cell bank. In this study, we choose three methods to detect the mycoplasma contamination of 100 kinds of the cell lines in our cell bank: DNA staining, direct method and PCR. By the comparison of different methods, we established the technique for Mycoplasma detection: using PCR kit to detect before storage, using three methods to detect fully after storage, using two methods to detect routinely, using PCR we established to detect in providing work.9 precious and rare cell lines were treated using BM-Cyclin and MC-210, and redetect the mycoplasma contamination after treatment for 1 week, 1 month and 1 year, repeating the test. According to the results, BM-Cyclin was selected as the first choice to remove mycoplasma contamination from the cell lines in cell bank.
