Diabetes is due to absolute or relative lack of insulin secretion, glucose metabolism disorder caused by high blood sugar as the main feature of the endocrine and metabolic diseases, is a kind of serious harm to human health, chronic disease, and its main clinical manifestations of polydipsia, polyphagia, urinary symptoms. In recent years, as people’s living standards continue to improve and accelerate the aging of the population, the incidence of diabetes increased significantly due to various complications which has become one of the major diseases that threaten people’s lives. Compound Chinese medicine treatment of diabetes in China has a long history, and shows good results. According to traditional Chinese medicine treatment guidelines and current literature, we adopted the traditional Cuban side(Diabetes square) and its derivatives, such as multi-compound optimized combination of herbs, formulated to contain yellow, pure Chinese medicine altogether, TCS and other Chinese herbal medicines sugar granule formulation.Objective:Type 2 diabetic rat model to investigate the effects of new drugs sugar granule type 2diabetic rats hypoglycemic mechanism of action, provide the basis for further development of the laboratory sugar granule.Methods:SD rats were randomly divided into normal control group and diabetic model group.High-fat diet combined with low-dose intraperitoneal injection of streptozotocin(STZ) 28 mg ? kg-1 model of type 2 diabetes, after successful model rats were randomly divided into model group, high new Chinese medicine granule, medium and low dose group(22.8,11.4,5.7 g ? kg-1), metformin group. Normal control group and model group were given saline, new Chinese medicine granule each treatment group were orally administered dose accordingly. Gavage five weeks after fasting blood-glucose(fasting blood-glucose,FBG) changes; ELISA method to detect rat serum fasting insulin levels(fasting insulin levels, FINS), C- reactive protein( C react protein, CRP), changes in IL-6; the kit to detect rat serum triglyceride(triglyceride, TG), total cholesterol(total cholesterol, TC), high density lipoprotein cholesterol(high density lipoprotein cholesterol, HDL-C), low-density lipoprotein cholesterol(low density lipoprotein cholesterol, LDL-C), superoxide dismutase(superoxide dismutase, SOD), MDA(malondialdehyde, MDA) content; immunoblot assay of glucose in the liver RT- q PCR assay liver GK m RNA expression; expression of kinase(GK) protein.Results:(1) the use of high-fat diet combined with intraperitoneal injection of low-dose STZ-induced type 2 diabetes model is successful, the occurrence of insulin resistance.(2) compared with the normal group, FBG model group rats, FINS, IRI, TC, TG, LDL-C,CRP, IL-6, MDA levels were significantly increased(P <0.05,0.01), serum SOD significantly lower HDL-C content(P <0.01); 5 weeks after the administration of treatment,serum FBG hypoglycemic rats treated particles, IRI, FINS, TC, TG, CRP, IL-6 were significantly lower(P <0.05, 0.01), SOD levels were significantly increased(P <0.05,0.01);① serum LDL-C content of new Chinese medicine granule high dose group of rats significantly increased(P <0.01). HDL-C was increased, MDA content decreased(P> 0.05),no significant difference; significantly increased serum LDL-C content ② new Chinese medicine granule dose group(P <0.01), MDA was significantly decreased(P <0.01).HDL-C level was increased(P> 0.05), no significant difference. ③ increased serum levels of HDL-C particles hypoglycemic low dose group(P <0.01), LDL-C, MDA content decreased(P> 0.05), no significant difference.(3) New Chinese medicine granule can alleviate lipid metabolism in rats with type 2diabetes, inhibit the deposition of fat in the liver.(4) After the new Chinese medicine granule treatment, the expression of hepatic GKdiabetic rats increased, immunoblotting and RT- q PCR trends were similar.Conclusions:The new particles can significantly reduce blood glucose in type 2 diabetic rats, insulin resistance, the protective effect of the liver. The mechanism may relieve inflammation,improve the ability to oxidative stress in diabetic rats; raised liver glucokinase expression,inhibition of glycogen output related.
