Study on Mechanism of Anti-CD45RB mAb in Combination with Anti-CD154 mAb and DSG in Treating Islet Transplantation Objective:Adopting anti-CD45RB mAb combining with anti-CD 154 mAb and DSG to explore influence of islet transplantation on survival in diabetic rat.Methods:Diabetic model was established by injection of C57B/6 mice streptozotocin preparations. After the Sprague-Dawley mice’s pancreas was removed, islet cells were isolated and obtained by collagenase digestion method, which were then transplanted under kidney capsule of diabetic mice. Immunosuppressants were injected regularly by groups. Blood sugar, weight and glucose tolerance changes were observed to evaluate productivity.Results:High levels of blood sugar were found in mice from control groups 13-17 days after islet transplantation, and mice from DSG group became hyperglycemic 17-20 days after islet transplantation. High levels of blood sugar was found in one mouse on 92th day and 95th day respectively in anti-CD45RB mAb combining with anti-CD 154 mAb group, whereas no rat was hyperglycemic in anti-CD45RB mAb combining with anti-CD 154 mAb and DSG group for more than 100 days, with normal body weight increase.Conclusion:Anti-CD45RB mAb combining with anti-CD 154 mAb and DSG may prolong survival and life quality of diabetic mice with islet transplantation significantly.
