This paper explored the ultrasonic perforation effect for cells surface-enhanced Raman spectroscopy. In view of the insufficiencies of delivering nanoparticles into living cells by passive uptake method, a simple method based on ultrasonic perforation effect is put forward for the first time. It is to rapidly (1 minute) transfer the silver and gold nanoparticles (NPs) into living cells and use them as SERS enhancement within the basement by ultrasonic perforation effect, with an intention to carry out the study of the biochemical composition of cells and screening application of cancer cells. The main research results are to go as follows:1. Metal (silver and gold) nanoparticles inside a cell were observed by transmission electron microscopy (TEM) after ultrasonic perforation effect and SERS spectrums were successfully obtained from K562, NP69 and C666 cell lines using the ultrasound-mediated method for metal nanoparticles delivery in less than one minute;2. Through analysis of cell ultrasonic perforated-SERS spectra, the best optimized test conditions were achieved:using 20 times objective lens to expande the light beam to 100%;3. The whole cell SERS spectra obtained through ultrasonic perforation effect differ greatly from that achieved by traditional incubation using cell endocytosis method, it is probably caused by the toxic effects of silver nanoparticles;4. Getting SERS spectra of the NP69 cells and C666 cells and conducting a research on the screening of cancer cells by ultrasonic perforated effect-SERS technology. Then, using the same method to compare and analyze the NP69 cells and the leukemia K562 cells; Moreover, screening and analyzing the three kinds of cells by ultrasonic mediated SERS method.As a result, the ultrasonic perforation effect-SERS technique can quickly import the gold and silver nanoparticles that can be used as SERS substrate, into living cells. Through the testing SERS spectra, the quick recognition of biochemical components of cells can be achieved. This technique can also be applied to the screening of cancer cells. Actually, this method is not only simple and accurate, but also expand the application of SERS technique in the field of cell biological research, therefore, its application prospect is quite wide.
