Objective:AMP-activated protein kinase protein(AMPK) is an important energy homeostasis in cellular. It has been reported that AMPK is low in metabolic diseases, such as obesity, type II diabetes. AMPK can regulate energy matebolism in cellular, and can also induce tumor cells apoptosis and inhibit the growth of tumor cells.Fatty acid synthase(FASN) is an important enzyme in the de novo synthesis fatty acid in the cells and can regulate the energy metabolism in cellular. FASN was highly expressed in liver,adipose tissue in cellular and low or undetectable levels in other tissues. More and more studies found that FASN is significantly upregulated in some common human tumor tissues and its avtivity was enhanced, and seriously affected cancer prognosis. Oxidative stress,the body produces excessive active oxygen but impaires intracellular antioxidant defense system, resulting in toxic effect on cells. Breast cancer patients through a variety of mechanisms can activate the oxidation reaction, so that the body enter the state of oxidative stress, have adverse effects on the prognosis of the patients.Breast cancer is one of the most common female malignant tumor, diabetes is also an important disease affecting women’s health, studies have found that people with diabetes, women were more likely to develop breast cancer, and breast cancer patients with type 2 diabetes were independent prognostic risk factors of recurrence and metastasis of breast cancer. The study on the expression of AMPKa, FASN protein and the analysis of oxidative stress in breast cancer combined with diabetes is less. The objective of the experiment is to measure AMPKa、 FASN protein and analyse the realitionship between AMPKa, FASN protein and clinical pathological characteristic. To explore its possible role in type 2 diabetes patients with breast carcinoma through the analysis of the content of SOD, MDA, GSH in serum in the controll 、the diabetes 、the breast cancer、the type 2 diabetes mellitus with breast cancer patients.Methods:Immunohistochemistric method was used to detect the expression of AMPKa and FASN protein in 30 cases breast cancer and 30 cases breast cancer combined with diabetes、30 diabetes and 30 health people were rolled in the study. The content of SOD、MDA、GSH were measured in serum.Results:1 AMPKa protein was significantly higher in breast cancer than that in breast cancer combined with diabetes mellitus, but FASN protein was significantly lower in breast cancer than that in breast cancer combined with diabetes mellitus(P<0.05)2 The expression of AMPKa、FASN were positively correlated with differentiation degree,lymph node metastasis, TNM stage(P<0.01).Correlation was not found between AMPKa、FASN and age(P >0.05)3 The comparison of oxidative stress(SOD、MDA、GSH) in serum in the controll 、the diabetes, the breast cancer and the breast cancer combined with diabetes mellitus3.1 Compared with the healthy group, SOD 、GSH was decreased in the breast cancer,the diabetes group, the breast cancer combined with diabetes mellitus,and the difference was statistically significant(P<0.01);Compared with the breast cancer combined with diabetes mellitus,the content of SOD、GSH was increased in the breast cancer,the diabetes,and the difference was statistically significant(P<0.01);Despite the content of SOD、GSH in serum in the breast cancer and the diabetes was not the same,the difference was not statistically significant(P>0.05).3.2 Compared with healthy group, MDA was increased in the breast cancer,the diabetes group, the breast cancer combined with diabetes mellitus,and the difference was statistically significant(P<0.01);Compared with the breast cancer combined with diabetes mellitus,the content of MDA was decreased in the breast cancer,the diabetes,and the difference was statistically significant(P<0.01);Despite the content of MDA in serum in the breast cancer and the diabetes was not the same,the difference was not statistically significant(P>0.05).4 The comparison of blood lipid in serum in the controll 、the diabetes, the breast cancer the breast cancer combined with diabetes mellitus4.1 Compared with the healthy group, TG、TCH was increased in the breast cancer,the diabetes group, the breast cancer combined with diabetes mellitus,and the difference was statistically significant(P<0.01);Compared with the breast cancer combined with diabetes mellitus,the content of TG was decreased in the breast cancer,the diabetes,and the difference was statistically significant(P<0.01);Compared with the breast cancer combined with diabetes mellitus,the content of TCH was decreased in the breast cancer and the diabetes,but the difference was not statistically significant(P>0.05);Despite the content of TG、TCH in serum in the breast cancer and the diabetes was not the same,the difference was not statistically significant(P>0.05).4.2 Compared with the healthy group,FASN was increased in the breast cancer,the diabetes group, the breast cancer combined with diabetes mellitus,and the difference was statistically significant(P<0.01);Compared with the breast cancer combined with diabetes mellitus,the content of FASN was decreased in the breast cancer,the diabetes,and the difference was statistically significant(P<0.01);Despite the content of FASN in serum in the breast cancer and the diabetes was not the same,the difference was not statistically significant(P>0.05).4.3 Compared with healthy group, NFEA was increased in the breast cancer,the diabetes group, the breast cancer combined with diabetes mellitus,and the difference was statistically significant(P<0.01);Compared with the breast cancer combined with diabetes mellitus,the content of NFEA was decreased in the breast cancer,the diabetes,and the difference was statistically significant(P<0.01);Despite the content of NFEA in serum in the breast cancer and the diabetes was not the same,the difference was not statistically significant(P>0.05).Conclusions:1 The expression of AMPKa protein was decreased and the expression of FASN protein was increased in breast cancer combined with diabetes;The expression of AMPKa、FASN were positively correlated with differentiation degree,lymph node metastasis, TNM stage.AMPKa、FASN protein may be markers of tumor prognosis.2 The content of SOD、GSH was decreased and MDA was increased in patients with breast cancer combined with diabetes.The content of TG、TCH、FASN、NFEA was increased in patients with breast cancer combined with diabetes. oxidative stress and lipid metabolism disorders may be one of the mechanisms leading to type 2 diabetes mellitus complicated with breast cancer.
