Objective To explore the role of scavenger receptor in macrophage response initiated by wear debris of the orthopedic artificial materials.Methods Mouse peritoneal macrophages were isolated and cultured until adherent to the plate. Part of the well of the peritoneal macrophages were pretreated with certain concentration of polyguanylic acid, which has the ability to blcok scavenger receptor,before all of the well stimulated with certain concentration of polymethylmethacrylate (PMMA) particles,particles of TiAl6V4 (TiPs) and CoCrMo (CoPs).The supernatant was collected at certain point of time and the content of TNF-a in the supernatant was measured by ELISA.The content of TNF-a in the culture medium which were co-incubated with poly(G) was compared by which not. The analysis We applicated SPSS 13.0 statistical software to analysis the data, using independent samples t-test to test the hypothesis. P<0.05 was considered statistically significant.Results Our ELISA results showed that no matter what kind of particles,it can stimulate macrophages to produce TNF-a significantly higher than the control group (P<0.05).And the induction of polyguanylic acid significantly decresed the level of TNF-a for TiPs and CoPs.which proved by the P-value which is less than 0.05.However,for PMMA particles,whether the addition of poly(G) did nothing with the release of TNF-a,which is also proved by P-value which is greater than 0.05.Conclusion In this experiment,we validated that scavenger receptor may play a role in the macrophage response caused by some kind of wear particles,but the specific role is unknown.There is a lot of work to be done as a breakthrough to treat periprosthetic osteolysis in patients with hip replacements.
