Water extraction method to extract Sparassis crispa crude polysaccharide. Decolorization and deproteinizing, separation and purification of polysaccharides, measuring their monosaccharide composition and infrared spectroscopy. The results showed that D301T ion exchange resin decolorization rate reached 81.60%,it was the best resin to Sparassis crispa polysaccharide decoloring. Separating the two main components of Sparassis crispa polysaccharide, Distilled water eluted fraction(SCP1) and 0.1% NaCl eluted fraction(SCP2), SCP1 and SCP2 monosaccharide composition are mannose (Man), glucose (Glu), galactose (Gla) and fucose (Fuc), the mole ratio were 0.043:0.06:1.75:0.386 and 0.0554:1.656:0.1249:0.0201. Infrared spectroscopy analysis Sparassis crispa polysaccharide which has been the characterized by structural polysaccharidesUsing the chemical oxidation in vitro experiment, Establishment of aging model, fed different concentrations of Sparassis crispa polysaccharide (SCP) in vivo antioxidant experiment. The results showed that Sparassis crispa polysaccharide SCP、SCP1 and SCP2 on hydroxyl radicals, ABTS and DPPH had a better scavenging effect,they also had good reducing power and and SCP2 effect is the most best. In vivo experiment, SCP Reduce the content of MDA in mice,and it also increased the content of SOD and CAT in mice.Using MTT method, the inverted microscope observation and fluorescent staining of Sparassis crispa polysaccharides inhibited the proliferation of human hepatoma HepG-2 cells. The results showed that Sparassis crispa polysaccharides on human hepatoma HepG-2 cells had better inhibition, SCP2 effect is the most best, the inhibition effect of 24h and 48h were respectively 51.12% and 53.45%. Inverted microscope method and fluorescent staining can observe Sparassis crispa polysaccharides had make the apoptosis of human hepatocellular carcinoma HepG-2 cells.Construction of with S180 cell tumor model in mice,fed different concentrations of Sparassis crispa polysaccharide(SCP), determination of mice tumor size, spleen and thymus index, serum level of TNF-α与 TNF-β content.tumor tissues were Observed by paraffin sections, the mice were survival test. Sparassis crispa polysaccharide had obvious inhibition effect on tumor in mice, the high dose group of Sparassis crispa polysaccharide inhibition rate was 50%, it improved the spleen and thymus index, and increasing the content of mice TNF-α-与TNF-β. Paraffin section observation of tumor tissue indicated the characteristics of apoptosis and pathological characteristics. Sparassis crispa polysaccharide could prolong the mice life and the high dose group could prolong the the life of mice was 32.6%.This study shows that, Sparassis crispa polysaccharide with high biological activity, SCP1 purified polysaccharide which is galactose-based homogeneous polysaccharide, SCP2 is a glucose-based homogeneous polysaccharide, Glucose-based component of the biological activity of the highest SCP2, and had good antioxidant, In vitro inhibited the proliferation of human hepatoma HepG-2 cells is also the best. Sparassis crispa polysaccharide in vivo could inhibited mouse S180 of tumor and prolonged survival of tumor mice life antitumor and antioxidation effect, It is worthy of attention and development of medicinal fungi.
