Sinularia sp. belongs to Family Alcyoniidae, Suborder Alcyoniina, Order Alcyonacea, Subclass Octocorallia, Class Anthozoa, Phylum Cnidaria. From the first time to research it in 1975, it is confirmed that about 50 species of the genus Sinularia have been studied, many steroids, terpenoids, alkaloids and polyketides have been discovered, showing that the genus Sinularia has rich structural and biological diversities. In particular, in recent years, cyclopentene ketone, sulfur-containing alkaloids, three acetyl glycolipids, chlorinated norcembranoids which are more unusual and more active are isolated from Sinularia sp., showing inherent complex forms and chemical diversity of SinulariaThere are rich natural resources and the largest amount of islands in Paracel Islands located in the northwest of the South China Sea. Its coral reef ecosystem distributes with all kinds of reef species for the special geographical position of Paracel islands. In the process of collecting samples, we found large benthic animals represented by soft corals are widely distributed in Paracel islands especially some rare coral species, at the same time, due to its complete and original ecological environment and metabolic pathways, it makes it possible for them to produce novelly active secondary metabolites. In order to find natural products with novel structure, pronounced biological activities and medical value, we have performed a systematic investigation on the chemical components and biological activities of the Sinularia sp. collected from Paracel Islands of South China Sea in 2012.Isolation and structure determination:The MeOH-CH2Cl2 extraction of the Sinularia sp. was chromatographed on TLC, Sephadex LH-20, silica gel and semi-preparative HPLC, respectively. Twenty-four compounds were identified by spectral methods (NMR、MS), the physical and chemical properties, there are cinanthrenol A (ZA-1), apo-9’-fucoxanthinone (ZA-2), one cyclopentenone derivative: suberosanone B, suberosanone A, hydroxydihydrobovolide, sinularone C (ZA-3-ZA-6), one sesquiterpenoid (ZA-7), seven diterpenes derivatives (ZA-8-ZA-14), five polyhydroxy sterols (ZA-15-ZA-19), two benzene derivatives (ZA-20-ZA-21), one pyrimidines (ZA-22), two nucleosides(ZA-23-ZA-24).Biological activities:The crude extract was evaluated for their cytotoxicities against HL-60 (human leukemia cells) and K562 (chronic myelogenous leukemia cell lines of the original) cancer cell lines by the MTT and SRB methods with the doxorubicin as a positive control. Most pure compounds were evaluated for their cytotoxicities against K562 and Jurkat The results show that compound 11 showed strong cytotoxic activity against Jurkat cell lines with IC50 values of 8.97μM.
