Tooth formation is the result of the interaction between epithelial and mesenchymal. so build a bioengineered teeth also need epithelial and mesenchymal tissue together. In 2000, Gronthos put human dental pulp stem cells in conjunction with HA/TCP powder which transplanted into immunocompromised mice, then they generated a dentin pulp-like structure. In addition, in 2010, Wang recombined the E13.5 dental mesenchym with human keratinocytes which was isolated from the pediatric foreskin, then the recombinants can form a complete teeth. The study found that the Rhesus monkey dental pulp stem cells and keratinocytes stem cells which act as the seed cells of tooth regeneration will draw people’s attention.Rhesus monkeys is similar with people on the evolution. It is the good choice of animal models from basic research to clinical research. Though Rong Lu has been successfully separated rhesus monkey keratinocyte stem cells and Huan had also been successfully separated rhesus monkey dental pulp stem cells. But the research of rhesus monkey keratinocyte stem cells and dental pulp stem cells which used in tooth regeneration are sarely report. So the odontogenetic differentiation research of rhesus monkey dental pulp stem cells and rhesus monkey keratinocyte stem cells have a great significance.This study selected a rhesus monkey tooth germ period of 7-8 weeks,9-10 weeks, 11-12 weeks as the research object, after HE staining to observe its morphological structure. The results showed that 7-8 weeks of tooth germ were in the transition period of bud stage to cap stage,9-10 weeks of tooth germ were at the bell stage,11-12 weeks of tooth germ were at the differentiation stage. Meanwhile we detected the expression of PITX2, FGF8, MSX1 at this stage in Rhesus monkeys’tooth germ. The results showed that the PITX2, FGF8, MSX1 in these times of rhesus monkeys tooth germ have expressed more or less.This study successfully separated rhesus monkeys keratinocyte stem cells, and recombined the E13.5 dental mesenchym with the second generation of rhesus monkeys keratinocyte stem cells, then the recombinants form a complete biological function of teeth. At the same time, we also successfully separated rhesus monkeys dental pulp stem cells, and confirmed that it had adipogenic osteoplastic and dental differentiating potential. These proved that rhesus monkeys keratinocytes stem cells and rhesus monkeys dental pulp stem cells can be used as seed cells to realize tooth regeneration in the future. It is important for achieve to tooth regeneration from basic research to clinical research by using stem cells technology.
