Asterias rollestoni, a genus of Echinodermata animal phyla. Asteroidea, forcipulatida asterid families, is one of the common marine macroinvertebrates. Asterias rollestoni was originally contained in the Compendium of Mater ia Medical. Medicinal value includes its significant pesticide effect on stomach and liver calming and, analgesic efficacy as well as its ability to stop acid producing.To establish a comprehensive components group, the extraction solvent is respectively selected purified water and 95% ethanol and the ultrasonic cleaning instrument was assisted as extraction in this experiment. The 95% ethanol extract were dispersed in water and successively extracted by petroleum ether, ethyl acetate, butanol solvent to obtain distribution component, in which the petroleum ether fraction, and the ethyl acetate components constitute a single component of Asterias rollestoni with small polar compounds in aqueous solution as a single component. Four components had been obtained in this step. Butanol fractions sample was separated by macroporous resin column and the solution of ethanol water was 20,40, 60,80,100%. The compound compositions of 80 and 100% ethanol elution fractions were more rich. Five components had been obtained in this step. Then the 80 and 100% ethanol elution fraction was separated by pressurized in silica gel column to get five silica gel column fractions respectively. Ten components had been obtained in this step. Above all,19 components had been obtained by 95% ethanol extract.17 components were obtained by purified water ethanol. The above 36 components constitute the Asterias rollestoni standard component. They established basis for separating characteristic compounds, identifying the structure and exploring the activities.To do more researches to explore their medical value, All components groups were tested by TLC to look for the new color points.On the basis of 60% groups separated by silica gel column, a simple component, presenting single color point as a flow rate of 0.66 in TLC and presenting two chromatograph peaks in HPLC chromatograph map, was obtained by normal phase silica gel, MCI gel, reversed-phase silica gel and gel column chromatography. On the basis of 80% groups separated by silica gel column, two alkaloid compounds were obtained by normal phase silica gel, reversed-phase silica gel and gel column chromatography:(3R,6S)- 3,6-bis((1H-indol-3-yl)methyl)piperazine-2,5-dione(1) and N-(2-(1H-indol-3-yl)ethyl)-2-phenylethanamine(2). Anti tumor activity test had shown that S4 could inhibit all six kinds tumor and it’s worthy of intensive study.In summary, the scientific standard components groups of Asterias rollestoni had been established at the first time by ultrasonic extraction, solvent partitioning, column separation. And their chemical compositions were presented by HPLC. On the basis of groups, S4 was obtained which consisted simple substance and two alkaloid compounds were separated. As a rich source with a wide range of biological activity of marine organisms, Asterias rollestoni is easy to obtain and has important medical value, which makes in-depth study of the material base of it a great significance.
