In recently years, tumor probability has become more increasingly prominent, and the methods of treating cancer have become been increasingly diversified. The traditional therapy: chemotherapy is currently the most widely used treatment, but its less effectiveness and side effects troubles in the clinical treatment. And Photodynamic therapy is a new means of treating cancer, which attracts more and more attentions, due to its validity and uniqueness. but also it faces a huge challenge, because the photosensitizers which can produces singlet oxygen, are mostly hydrophobic, difficult to arrive at the tumor. It is necessary and key to build a safe and can carry out a variety of different mechanisms of drugs. Combining two treatment, it can further enhance the effect of cancer.In this paper, I constructed the poly-lysine modified graphene(G-PLL), while carrying a chemotherapy drug doxorubicin(DOX) and a photosensitive drug zinc phthalocyanine(ZnPc), combining chemotherapy and photodynamic therapy to inhibit the growth of various tumor cells G-PLL carrier has good biocompatibility and stability: the size of it is about 200 nm and Zeta potential of up to +29.25 mV. G-PLL can carry alone doxorubicin(drug loading:110%) and photosensitizer zinc phthalocyanine(drug loading:59%).Further it is capable of carrying two drugs simultaneously to form a nanocomposite G-PLL/DOX/ZnPc which has good stability and dispersibility. Through the biological study, under irradiation at a wavelength of 670 nm, it can produce singlet oxygen successfully. The results showed that, nanocomposite can be swallowed in the variety tumor cells. Based on the inhibition rate of 50% CI50 synergy index were 0.07(HeLa),0.13(MCF-7) and 0.13(B16), the nanocomposite has strong chemotherapy and photodynamic therapy synergie effect in multiple tumor cells.This study shows that the drug G-PLL/DOX/ZnPc nanocomposite has a good synergistic inhibition of tumor cell proliferation and promote apoptosis of tumor cells, offering a new way of cancer treatment for cancer combination therapy.
