Objective Investigating the expressions, the cellular localizations of Triad1 and UBE2Q1 after rat traumatic brain injury. To further confirm the interactions between Triad1 and UBE2Q1, and its significance in nerve repair after injury.Methods Sprague-Dawley(SD) rats were subjected to brain injury, Expression patterns and distribution for Triad1 and UBE2Q1 were observed in the injured cortex by Western Blot, immunohistochemistry, RT-PCR and immunofluorescence. Subsequently, double immunofluorescence labeling and co-immunoprecipitation were performed to study the interactions between Triad1 and UBE2Q1, and to explore their pathological and physiological significance during nerve injury and regeneration. Build a H2O2 stimulated cells model, knocking-down or over-expression of Triad1 and UBE2Q1 and using flow cytometry certify the functions of Triad1 and UBE2Q1 after traumatic brain injury.Results Investigate the expression profiles of Triad1 and UBE2Q1 in adult rat brain cortex after traumatic brain injury through western blot and RT-PCR. The mainly co-localization of Triad1/UBE2Q1 with neuronal marker by double immunofluorescent staining, but the astrocytes marker is few. Triad1 interacts and co-locates with UBE2Q1 in CNS. Detection of neuronal apoptosis and relative Triad1 and UBE2Q1 changes in adult rat brain cortex after TBI.Meanwhile, some pro-apoptosis factors, such as p53、Bax、active caspase-3, have high expression after injury. Neuronal apoptosis may be dependent on the level of expression of the p53 after TBI.Conclusion 1 TBI induced the changes expression of Triad1 and UBE2Q1,and they interacted with each other;2 The interactions between Triad1 and UBE2Q1 are involved in neuronal apoptosis; 3 Interactions between Triad1 and UBE2Q1 are involved in central nervous system(CNS) injury and repair in cell level;4 Further certify Triad1 is one of the E3 ubiquitin ligases and has the activity in neural central system.
