Objective:To explore the effect of bufalin apoptosis and autophagy of human bladder cancer T24 cells,and study the mechanism of bufalin disturbance growth of bladder tumor T24 cells.Methods:Using T24 cells as the object of study of bladder cancer.Different concentrations of bufalin for bladder cancer T24 cells.The proliferation-inhibiting effect of Cino on T24 cells were measured by methyl- thiazole tetrazolium(MTT)colorimetric assay.The cell morphology of culture cells were observed under the inverted microscope after 24 hours.The cell cycle effects of bufalin on T24 cells were detected by flow cytometry analysis. The observation of autophagic vacuoles occurred by electron microscopic.The expressing of Beclin1 were tested by Real-time FQ-PCR.Results: The MTT results are shown:The IC50(half maximal inhibitory concentration) values were 91.7nmol/Land 20.4noml/L for respectively after different ncentrations bufalin acted on the human bladder cancer T24 cells in 24 hours and 48 hours.Bufalin can inhibit the proliferation of bladder cancer T24 cells.Flow cytometry data are indicated: The human bladder cancer T24 cells treated by bufalin,G0/G1 phase cell count than normal bladder cancer cells significantly increased, S phase cells of bladder cancer compared with normal bladder cell count was significantly decreased,results indicated that bufalin can’t make part of T24 cells into S phase, but the human bladder cancer T24 cells hinder the arrest in the G0/G1 phase, the cell proliferation was inhibited.Transmission electron microscope showed: bladder cancer T24 cells treated by bufalin,induced autophagy bubble generation.The m RNA levels of Beclin1 in T24 cells was markedly up-regulated after treatment with.Conclusion: Bufalin can significantly inhibited the proliferation of T24 cell, influence of cell cycle, inducing autophagy and apoptosis of the cells, may be related to the enhanced expression of autophagy related gene Beclin1 levels.
