Objectives: T o investigate the possible role and clinical significance of Shh and Gli1 genes in the pathogenesis of multiple myeloma.Hedgehog signal channel play a important role in the tumorigenesis,development,infiltration,metastasis of malignant tumor and resistance of pharmacotherapy.There are three Hedgehog genes in the vertebrates as follows:Sonic Hedgehog(Shh),Desert Hedgehog(Dhh),India Hedgehog(Ihh),encoding Shh,Dhh,Ihh protein respectively.Shh gene is very conservative,getting the widest research among the Hedgehog genes.Shh gene reside in the nervous system,bone, skin, external genitalia,extremities,hematopoietic cell and vascular endothelial cell etc,not only participating in the formation and maintenance of nervous system mesophragma, auxano-bone,formation of hematopoietic cell and vascular endothelial cell,but also regulating asymmetric physical development and morphogenesis of many organs including eyes,external genitalia,hair,pancreas,bone,lung,hypothalamus,teeth,spinal cord.As the nuclear factor of Hedgehog signal channel,localization in th nucleus and cytolymph,Gli transmits transduction signal to endonuclear.There are three Gli nucleus open gene in the mankind,including Gli1,Gli2,Gli3,in which there are C terminal transcriptional activation domain,meanwhile there are N terminal transcription inhibition domain except for Gli1.Gli1 mainly transcribe promoter,but Gli2 and Gli3 transcribe both promoter and repressor.In this study,we detect the expression of Shh and Gli1 gene by RT-PCR and Western Blot in the patient of multiple myeloma and normal control,and investigate the possible role of Shh and Gli1 genes in the pathogenesis of multiple myeloma,in order to provide new method for diagnosis and treatment.Methods: 1.Choose 30 cases of new diagnosed patients of multiple myeloma as test team from December in 2012 to May in 2014,and 20 cases of patients of non-hematologic malignancies as normal control from July in 2012 to May in 2014. 2.Extract 2ml bone marrow,anticoagulation by EDTA-2K,and separate mononuclearcell by Lydroxypropylmethyl Cellulose to prepare mononuclearcell suspension,storage in the-80℃ refrigerator.3.Extract total RNA of mononuclearcell,and compute the purity of specimen by spectrophotometer, reverse transcribe to c DNA then polymerase chain reaction amplification.4.Detect the expression of Shh and Gli1 protein by Western Blot.Results: The expression of Shh and Gli1 increased in the new diagnosed patients of multiple myeloma,compared with normal control.The expression of Shh and Gli1 increased in phase Ⅲ relevant with many clinical index,compared with phase ⅠorⅡ.There are no obvious difference between group A and group B.Conclusion: The expression of Shh and Gli1 increased in the new diagnosed patients of multiple myeloma,compared with normal control,relevant with many clinical index and Staging,which have something to do with the tumorigenesis and development of multiple myeloma.
