Objective: Preparation of the model mice of dysbacteria caused by antibiotic, using the related technology of modern animal experiment, through the comprehensive analysis on the general situation of mice, spleen index, thymus index and the content of s Ig A in intestinal mucosa, preliminarily studying the effect of Yingxie particles on immune function of model mice.Methods: 50 Healthy SPF Kunming mice(male and female) were randomly divided into normal group with 9 mice, model group with 41 mice, consulted and improved the modeling methods described in the related literatures, given Lincomycin Hydrochloride Injection to these mice by intragastric administration to prepare the mice model with dysbacteriosis and diarrhea. After the success of modeling, the model group was randomly divided into five groups, where one group was the blank model group, the other four groups were treatment groups. The normal group and blank model group were given saline for 5 days, the treatment groups were respectively given Chang Le Kang, Yingxie particles in high, middle and low dose for 5 days. After the treatment, sterilely collected fresh feces for inspection, sacrificed the mice after weighing the weight, then separated spleen and thymus, calculated spleen index and thymus index, and cutted the terminal ileum, then detected the content of s Ig A in intestinal mucosa of mice by ELISA method.Results: 1 The weight of mice: after the modeling, model group mice appeared diarrhea, red anal, and even rectal prolapse, poor mental reactions, diet decreased, lazy and so on, besides, the weight variation of mice decreased compared to the normal group(P<0.05);after the treatment, compared with the normal group, blank model group and Chang Le Kang group had statistical significance(P<0.05);compared with the blank model group, the normal group, Yingxie particlesin high, middle and low dose group had statistical significance(P<0.05); compared with Chang Le Kang group, Yingxie particles in high, middle and low dose group had statistical significance(P<0.05); and there was no significance among Yingxie particles in different dose(P>0.05).2 The results of spleen index of mice in each group: Compared with the blank model group, normal group、Yingxie particles in high, middle, low dose group had statistical significance(P<0.05); compared with Chang Le Kang group, Yingxie particles in middle and low dose group were better than that of Chang Le Kang group, there was a statistical significance(P<0.05); Yingxie particles in middle dosage group was better than normal group, there was a statistical significance(P<0.05); and there was no difference among Yingxie particles in high, middle, low dose group(P>0.05). 3 The results of thymus index of mice: the thymus index of mice in blank model group was lower than that of normal group、Yingxie particles in high, medium, low dose group(P<0.05); the thymus index of mice in Yingxie particles in high, middle dose group was higher than that of Chang Le Kang group, there was a statistical significance(P<0.05); the thymus index of mice in Yingxie particles in middle dose group was higher than that of normal group, there was a statistical significance(P<0.05); there was no difference among Yingxie particles of different dose group(P>0.05). 4 The content of s Ig A of mice intestinal mucosa in each group: compared with the blank model group, normal group, Yingxie particles in high, middle, low dose group had statistical significance(P<0.05); compared with the normal group, Chang Le Kang group、Yingxie particles in low dose group had statistical significance(P<0.05); compared with Chang Le Kang group, Yingxie particles in high, middle dose group had statistical significance(P<0.05); compared with Yingxie particles in middle dose group, Yingxie particles in high and low dose group had statistical significance(P<0.05);there was no difference between Yingxie particles in high and low dose group(P>0.05).Conclusion: 1. Antibiotics can cause the intestinal flora and low immune function of mice. 2 Yingxie particles in high, middle and low dose group can improve the general condition of dysbacteriosis model mice and improve the local intestinal and global immune function of the model mice. Among these three groups, the middle dose of Yingxie particles have the best curative effect, the next is Yingxie particles in high dose group, the last is Yingxie particles of low dose group. 3 Yingxie particles in different dose groups are better than Chang Le Kang group in improving the immune function of the modle mice with Dysbacteriosis and Diarrhea.
