Objective: Observation of the clinical curative effect and mechanism of Xianyuquan soluble granules, and impact on the contents of Aminobutyric acid,Glutamic acid and behavior of epileptic rats affected by pentetrazol,explore the new development of therapeutical methods of epilepsy to provide more basis for the application and spreading of Xianyuquan soluble granules. Methods: 70 SD rats aged 6~8 weeks and weighted 200±10g were randomized into a control group,a model group,a sodium valproate-treated group,a low-dose Xianyuquan soluble granules group and a high-dose Xianyuquan soluble granules group(14 cases in each group).While the rats in control group were fed with physiological saline in the dose of 37.5mg/kg?d by intragastrical perfusion,the other rats were fed with pentetrazol in the same dose and way. During the procedure of modeling,the low-dose group were administrated with 568 mg/kg?d and the high-dose group in the dose of 1172 mg/kg?d with Xianyuquan soluble granules,while the rats in sodium valproate-treated group with sodium valproate in a dose of 200 mg/kg?d for excessive twenty-eight days.At the same time,the control and model group rats were administrated with physiological saline in the dose of 1 ml.The detailed performance was recorded every day.Rats were executed on the 28 th day.The contents of GABA and GLU in the hippocampus of every rat were measured on HPLC-MS/MSQ-TRAP. The datas of the experiment were analyzed by the usage of software package of SPSS19.0.It is of high statistical sense when the P<0.05. Results:1.6 rats have not met the standard of kindling,and 3 rats died from excessive epileptic attack,and 2 rats were killed by the others in the same cage.so datas of 59 rats were statisticaliy analyzed.2.Compared with the model group,the psychic,fur color and activities of rats treated with medicine have been significantly improved.Compared with model rats,the weights of rats in model group have declined(P<0.05).While the weights of rats administrated with medicine decreased compared with model rats(P<0.05).And the gain weights of rats in high-dose are more than that of the low-dose and sodium valproate-treated group(P<0.05).The conclusion that medicine can meliorate the nutritional status of epileptic rats and the curative effect of high-dose Xianyuquan soluble granules is better than that of sodium valproate is reached.3.Compared with the model group,the epileptic latent period of rats anminiatrated with medicine was longer,while the lasting time of spasm was shorter(P<0.05).The differentiation of epileptic latency between sodium valproate-treated group,low-dose and high-dose group is of high statistical sense(P<0.05),and there was no obvious differenciation between the high-dose group and the low-dose group(P>0.05),and this result illustrates that Xianyuquan soluble granules and sodium valproate can prolong the epileptic latency in different degree and the effect of sodium valproate was better than that of Xianyuquan soluble granules.The differentiation of shortening the lasting time of spasm between the high-dose group and the sodium valproate-treated group is nonsense(P>0.05), but the effect of the two groups is better than that of the low-dose group(P<0.05),this result illustrates that sodium valproate and Xianyuquan soluble granules can shortening the lasting time of spasm in different degree and the effect of the two is nearly the same.4. The measurement of Glu in the hippocampus:The GLU in the hippocampus of model group have declined compared with that of the control group(P<0.05).The GLU in the hippocampus of rats administrated with medicine have that o declined compared with that of model group(P<0.05).There is no conspicious differenciation between the Xianyuquan soluble granules-treated group and sodium valproate-treated group(P>0.05),and there is no statistical sense(P<0.05).So the sodium valproate and Xianyuquan soluble granules can declined the contents of Glu in the hippocampus in the different degree,and no significant difference among the high-dose,low-dose and sodium valproate-treated groups was observed.5. The measurement of GABA in the hippocampus:The GABA in the hippocampus of model rats have declined compared with that of rats in the control group(P<0.05).The GABA in the hippocampus of rats administrated with medicine have increased compared with that of model group(P<0.05).There is no conspicious differentiation between the high-dose group and sodium valproate-treated group(P<0.05).So the GABA in the hippocampus can increase in different degree after the usage of sodium valproate and Xianyuquan soluble granules and the effect of the two kinds of medicine is nearly the same. Conclusion:1. Xianyuquan soluble granules can improve the general status of epileptic rats conspiciously,and can prolong the epileptic latent period and shorten the lasting time of spasm;2.The Xianyuquan soluble granules may decrease the contents of GABA and increase the content of Glu in the hippocampus while playing anti-epileptic effect.
