Objective: Parkinson’s disease(Parkinson ’s Disease, PD) is one of the most common in the elderly population of chronic, progressive neurodegenerative diseases, in older people have a higher prevalence and morbidity. Our existing PD patients more than 1 million, over 65 years population prevalence rate as high as 2%, and increased with the increase of age, is expected in 2030 will reach 5000000 of PD patients in china. PD in the early clinical mainly extrapyramidal movement damage is given priority to, main show is resting tremor, slowness of movement, increased muscle tone and postural reflex disorders, some patients have intellectual impairment even disabled, seriously affect the patient’s health and survival ability. The main feature of PD is the nigrostriatal pathway dysfunction, pathological manifestations of dopaminergic substantia nigra pars compacta neurons degeneration and necrosis with intracytoplasmic eosinophilic inclusion bodies- Louis(Lewy bodies, LB) formation.The pathogenesis of PD is still not clear at present, in theprocess of degeneration of dopaminergic neurons, research on the death mechanism, found in the substantia nigra pars compacta in PD patients and in animal models, in addition to a large number of dopamine neurons loss has been found, and the activation of astrocytes. Existing research results show that astrocytes have a role in the pathogenesis of PD. This topic uses 6-OHDA unilateral striatal stereotactic microinjection of preparation of rat PD rotation model, the effects of c Electroacupuncture on PD rat model behavior of counting, midbrain striatal Glu concentration and GFAP, Cx43 expression, to explore the effects of astrocytes in rat substantia nigra dopaminergic may the mechanism of neuronal injury in and acupuncture intervention, in order to open up a new way of thinking for the acupuncture treatment of PD.Methods: 140 healthy male SD rats, were randomly divided into normal group, sham operation group, model group, electro acupuncture group. normal group, sham operation group with 30 rats in each group, model group, electro acupuncture group with 40 rats in each group. By microinjection to right side of rat striatal injection of 6-hydroxydopamine preparation lateral rotation model of PD rats, sham operation group, normal saline 6- Ohda micro injection. The normal group, model group, sham operation hanges of TH positive neurons, to observe the effect of group without any treatment; acupuncture group after making the mold on the basis of electroacupuncture at "Fengfu" "Taichong" point, using a continuous wave, frequency of 2Hz, strength of 1m A, duration 30 min, once a day, for two weeks. After EA treatment,(1) the rotational behavior of rats was observed by the rotational behavior of tester,(2) The number of TH positive neurons by immunohistochemistry detection(3)Using the detectionof striatal Glu concentrations of HPLC fluorescence method(4), RT-PCR method was used to detect changes in striatum of rat GFAPm RNA, Cx43 m RNA expression,(5) Using the method of Western-blot expression changes of striatum of GFAP, Cx43 protein activity detection.Results:(1) Behavioral observation, compared with model group, EA group rotation behavior significantly improved(P<0.01).(2) Compared with the normal group, sham operation group, the number of TH positive neurons of model group decreased significantly(P<0.01); compared with the model group, electro acupuncture group, the number of TH positive neurons increased significantly(P<0.01).(3)Compared with the normal group, sham operation group, model group, the concentration of Glu increased significantly(P<0.01); compared with the model group, electro acupuncture group, Glu concentrations were significantly decreased(P<0.01).(4) Compared with the normal group, sham operation group, model group, the expression of GFAPm RNA Cx43 m RNA was significantly increased(P<0.01), compared with the model group, the expression of GFAPm RNA, Cx43 m RNA decreased significantly in EA group(P<0.01).(5) Compared with the normal group, sham operation group, model group, the expression of GFAP Cx43 protein was significantly increased(P<0.01), compared with the model group, electro acupuncture group, Cx43 protein expression of GFAP was significantly decreased(P<0.01).Conclusion: Acupuncture can improve the rotational behavior in PD rats effectively, increase the number of TH positive neurons, decrease the release of GLu, and the expression of GFAP and Cx43 protein can inhibit. Astrocyte activation is involved in theoccurrence and development of PD, and the mechanism might be mediated release of Glu; Electroacupuncture intervention can effectively inhibit the activation of astrocytes, decrease the release of Glu, the protection of dopaminergic neurons.
