Objective: To explore the immunomodulatory functions of Banxia Xiexin Decoction and its influences on the planting of transplanted BMSCs of ulcerative colitis(UC) rats model.Methods:①The optimized transplantation time of BMSCs determined: rats were randomly divided into control group, model group and transplantation group, and each was subdivided into 7-day group, 14-day group and 21-day group. The rat’s model was established with TNBS; whole bone marrow adherence method was used for BMSCs culture and evaluation; transplantation was performed through rats’ tail intravenous injection. General conditions of rats, CMDI scores of colon tissue and tested IL-4 and IFN-γ levels in rats serum with ELISA were observed. ②Influences of Banxia Xiexin Decoction immunomodulatory functions on BMSCs transplantation treating UC as well as the mechanism investigated: rats were randomly divided into normal group, model group, SASP group, Banxia Xiexin Decoction group, BMSCs group and Banxia Xiexin Decoction + BMSCs group. TDI scores of rats colon tissue, levels of IL-6 and IL-10 in serum, and planting of transplanted cells in lesions and expressions of bFGF were observd. Results: ⑴In the experiments of determining optimized time for transplantation: compared to the control group, both model group and transplantation group had increased CMDI scors and the level of IFN-γ,decreased the level of IL-4. In the 14 th day, the transplantation group had lower CMDI scors and IFN-γ content than the model group and higher IL-4 content than the model group, with significant difference(P<0.05). ⑵In the experiments of exploring influences of Banxia Xiexin Decoction combined with BMSCs transplantation on UC rats model: ①Compared the normal group, the model group, SASP group, Banxia Xiexin Decoction group, BMSCs group and Banxia Xiexin Decoction + BMSCs group all had increased CMDI and TDI scores. Compared to other groups, the Banxia Xiexin Decoction + BMSCs group had significantly decreased CMDI and TID scores, with statistical significance(P<0.05). ②Compared to the control group, both the model group and the Banxia Xiexin Decoction group had decreased IL-6 levels. The model group, SASP group, Banxia Xiexin Decoction, BMSCs group and Banxia Xiexin Decoction + BMSCs group had increased IL-10 levels. Compared to the model group, all intervention groups had decreased IL-6 content and increased IL-10 levels, with difference of statistical significance(P<0.05). ③In the BMSCs group and the Banxia Xiexin Decoction + BMSCs group, BrdU positive cells could be seen with immunohistochemistry and there were more in lesions than the healthy mucosa. While the Banxia Xiexin Decoction + BMSCs group had more positive cells than the BMSCs group. ④bFGF expression could be seen in each group and the Banxia Xiexin Decoction + BMSCs group had the most positive cells.Conclusion:①The 14 th day is preliminarily determined as the optimized time for BMSCs transplantation for treating UC. ②Banxia Xiexin Decoction combined with BMSCs transplantation can effectively promote restoration of UC rats’ colonic mucosa. ③Banxia Xiexin Decoction can improve inflammatory conditions of UC rats through regulating relative immunologic factors levels, to create conditions for repair of ulcers. ④Banxia Xiexin Decoction can promote planting of BMSCs transplantation onto lesions and directed differentiation into colonic mucosa stem cells and then colonic epithelial cells. ⑤ Banxia Xiexin Decoction immunomodulatory function on rats can promote reconstruction of intestine microenvironment, facilitates bFGF expressions and vascular regeneration, and restore impared colonic mucosa, may become one of the clinical treatment methods for UC.
