To analyze the function of Yiqi Huoxue Recipe(gavage the irr adiated rats)in radiation induced lung injury, and then Elaborate it w ith three methods(RT-PCR、 Immuno histochemistry), observe mRNA and protein expression variation of TGF-β1、 TNF-a、 NOS3 and MTHFR in the tissue of radiation-induced lung injury, to analysis the particular mechanism of the injury from protein and gene levels.108 healthy adult rats were equally divided into three groups randomly: named separately as simple radiation group(model group, n=36), radiation and Chinese herbology group(Chinese herbology gro up, n=36), and blank control group(control group, n=36). Confirm th e radiation region under x ray-Simulation Localization System: the up border to the top of the right lung, the lower bound for the right-h and Diaphragm top, the left border is right-hand edge of the spinal c ord, right up to the right side of the thorax outer boundary. Radiatio n field is generally 3 × 3cm size. Occlusion of the left lung and me diastinal lead weights, totally 30 Gy, 5Gy / time, a total of six times.Rats of Model group and Chinese herbology group were irradiated with 6MV-X Linear Accelerator,control group rats just anesthesia, bu t not radiation.Before irradiation,the rats were anesthesiaed with 7%chloral hydrate aldehyde(5?kg-1) by intraperitoneal injection, then pro ne on the platform and fixed them(nail four nails on the plank, the l imbs of anesthetized rats fixed on the nail were rubber band). From the first day of radiation,rats of Model group and Chinese herbology group were fed with Yiqihuoxue prescription,1ml respectively, 2 times a day,at 08:00 and 20:00. Six rats were randomly selected at the end of 2,4,8,12,16,20 weeks, open the chests of these rats i mmediately right after intraperitoneal anesthesiaed, take the right lung tissue and save in right way. PCR methods were used to detect dyn amic changes of TGF-β1, TNF-a, NOS3, MTHFR mRNA expression at each time pointand immune histochemistry were used to detect the dynamic changes of TGF-β1, TNF-a, NOS3, MTHFR protein, and t he experimental datas were statistical analysised with SPSS.17.0.RT-PCR and immunohistochemistry results suggest that TGF-β1, TNF-a, NOS3, MTHFR mRNA and protein expression after irradiatio n among the 2th-20 th weekends appear a first increaseing and then do wnward trend. Their protein and mRNA expression began to increase from the 2th weekend, 8th weekend to the peak expression, decrease d from 12 th weekend. MRNA and protein’s expression trends of Chi nese herbologygroup were similar with the model group, but there ar e different variations at different time points.We found that TGF-β1, TNF-a, NOS3 and MTHFR genes were sensitive factors when radiation-induced lung injury occurs. Yiqihuoxu e can regulate the expression of TGF-β1, TNF-a, NOS3, MTHFR fro m transcription and translation level in radiation-induced lung injury at different time points. This may be the molecular mechanisms of d rug prevention and treatment in radiation-induced lung injury. After t he end of radiotherapy,relevant cytokines’ expression continued to 16 weekend, it suggested that the treatment time of radiation-induced lu ng injury should be extended to 2-3 months after the end of radiati on. And at different times, the priorities and the rules of treatment is different to be emphasesed.
