Objective:To compare the differences effect of bladder cancer cell lysate and cancer cell culture supernatant of T lymphocyte migration ability.To detect the expression and significance of T lymphocytes and cells of PEDF in peripheral anticoagulated blood of bladder cancer patients.To investigate the relationship between its and bladder cancer cells.To the early diagnosis, treatment and prognosis of bladder cancer.Methods:We selected 36 patients with untreated bladder cancer in the 1 st Department of Urology, the Second Affiliated Hospital of Kunming Medical University from December 2013 to November 2014 and 20 cases of healthy volunteers, their peripheral anticoagulated blood were collected;By the lymphocyte separation and counting technology, specific T cells in peripheral blood levels.Using transwell invasion experiment to compare the differences effect of bladder cancer cell lysate and cancer cell culture supernatant of T lymphocyte migration ability.The PEDF in different expression of T lymphocyte chemotaxis ability were determined at the level of mRNA by Real-time quantiative Polymerase Chain Reation(qRT-PCR) and Western Blot.Results:1.In peripheral blood of untreated bladder cancer patients, the expression level of T cells was significantly lower than that of healthy volunteers, the difference was statistically significant(p<0.05);2.Tumor cell culture supernatant and freeze the tumor cell lysate suspension of human peripheral blood T lymphocytes migrate chemotaxis and serum-free RPMI-1640 medium (control group), the difference has statistics migration significant (all P<0.05), tumor cell culture supernatant of chemokines stronger than freezing and thawing tumor cell lysate suspension (all P<0.05); and in a certain time frame, the tumor cell cultures Migration serum chemotactic ability prolonged incubation time weakened, (1H culture supernatant group> 4H culture supernatant group> 8H culture supernatant group> tumor cell lysate group> 1640 group)Tumor cell supernatants and cell lysates thawing tumor suspension of peripheral blood T lymphocytes migrate chemotaxis compared to serum-free RPMI-1640 medium (control group), the difference was statistically significant (P<0.05), migration of tumor cell lysate suspension thaw chemotaxis stronger than tumor cell culture supernatant (all P<0.05); and in a certain time frame, the migration of tumor cell culture supernatant chemotaxis prolonged incubation time and weakened (tumor cell lysate group> 1640 group> 1H culture supernatant group> 4H culture supernatant group> 8H culture supernatant group).The supernatant of tumor cells and tumor cell lysates thawing suspension in healthy volunteers and in patients with bladder cancer, T lymphocyte migration chemotaxis effect comparison:(1) 1640:There were significant differences (P peripheral blood T lymphocytes mobility<0.05); bladder cancer patients> healthy volunteer group;(2) freezing and thawing tumor cell lysate suspension:the mobility of peripheral blood T lymphocytes were significantly different (P<0.05); bladder cancer patients> healthy volunteer group;(3) tumor cell culture supernatant 1H:the mobility of peripheral blood T lymphocytes was no significant difference (P> 0.05);(4) tumor cell culture supernatant 4H:There were significant differences (P<0.05) in peripheral blood T lymphocytes mobility; bladder cancer patients<healthy volunteer group;(5) tumor cell culture supernatant 8H:There were significant differences (P<0.05) in peripheral blood T lymphocytes mobility; bladder cancer patients<healthy volunteer group.3.PEDFmRNA expression in T cells was significantly lower in bladder cancer patients.In different culture conditions of T lymphocytes attack of PEDF gene expression is different,expression in 1640 groups is the strongest,8 hours on tumor cell culture supernatant express the weakest(1640> 1 h culture supernatant group> tumor cell cracking material group> 4 h culture supernatant group> 8 h culture supernatant group).PEDF protein expression in T cells was significantly lower in bladder cancer patients, In different culture conditions of T lymphocytes attack of PEDF gene expression is different,expression in 1640 groups is the strongest,8 hours on tumor cell culture supernatant express the weakest(1640> 1 h culture supernatant group> tumor cell cracking material group> 4 h culture supernatant group> 8 h culture supernatant group).Conclusion:1.The reduce of T lymphocyte in peripheral blood of bladder cancer patients may cause immune escape important role in the development of bladder cancer.The role of T lymphocyte in the tumor biology of bladder cancer might have potential therapeutic implications.2. In patients with bladder cancer, tumor cell migration thaw lysates of T lymphocyte chemotaxis was significantly stronger than the culture of a certain time of bladder tumor cell supernatants.3.PEDFmRNA and PEDF protein expression in T cells was significantly lower in bladder cancer patients.In different culture conditions of T lymphocytes attack of PEDF gene expression is different,expression in 1640 groups is the strongest,8 hours on tumor cell culture supernatant express the weakest(1640> 1 h culture supernatant group> tumor cell cracking material group> 4 h culture supernatant group> 8 h culture supernatant group).Speculated that PEDF gene may be involved in T lymphocyte proliferation, migration and antitumor immunity.
