Plasma MiRNA-30, MiRNA-199, MiRNA-208 And MiRNA-374 Application In The Diagnosis Of Acute Coronary Syndrome

Objective:miRNA mediated various pathophysiological processes of cells in cardiovascular system, such as the proliferation,migration, differentiation, apoptosis, and so on, and played an important role in the development of cardiovascular diseases by regulating the expression of gene, disease. We detect expressions of miRNA-30d,miRNA-199a,miRNA-208b and miRNA-374bin plasma of STEMI,NSTEMI,UA,SA patients and control group using real-time PCR and discuss the possibility of plasma miRNA as biological markers in acute myocardial infarction diagnosis.Methods:We collect plasma of 50 STEMI patients,50 NSTEMI patients,50 UA patients,50 SA patients and 30 healthy persons and extract RNA from the plasma using the common QIAzol method. miRNA were quantified by using quantitative reverse transcriptase-polymerase chain reaction(qRT-PCR) assay. Meanwhile,we detect CK-MB and cTnI in the plasma of each group and study their correlation with miRNA,and their sensitivity and specificity in the diagnosis of acute myocardial infarction.Results:Plasma miRNA shows higher expression in STEMI,NSTEMI,UA and SA patient than control group, with statistical significance (P<0.05). Meantime,miRNA expression levels have positively correlation with cTnI and CK-MB. And the ROC curve shows miRNA are superior to cTnI and CK-MB in the diagnosis of acute myocardial infarction,especially in patients with chest pain less than 3 hours.Conclusions:These results suggest that specific miRNA myocardial cells can be used as a new class of biomarkers for early diagnosis of acute myocardial infarction and the assessment of prognosis.
