Objective: The goal of this experiment was to establish Enterococcus faecalis infection in experimental periapical SD rats as experimental model, and study the expression of Bruton tyrosine kinase expression(Btk) mRNA in the process of SD rats periapical lesions, to investigate the effect of Enterococcus faecalis on the expression of Btk.Methods: 32 healthy adult male Sprague- Dawley(SD) rats were selected as the experimental object. Rats was anesthetized by intraperitoneal injection of 10% chloral hydrate(0.35-0.40ml/100 g weight), rats were divided randomly into A, B two groups, 16 for each groups. Selecting on the right mandibular first molar oeclusal for experiment, to open the rats medullary cavity.Group A(no bacterium group) : in the rat medullary cavity sealed sterile PBS buffer solution small cotton balls; Group B(bacterium group): placed with small cotton ball which enclosed in a certain concentration of bacterial suspension and PBS buffer. Then all the groups were sealed by imported glass ionomer cement to fill pulp cavity. The left side mandibular first molar of the rats without any treatment were regarded as the normal control group. Conventional uniform feeding in cages. After that, A, B two experimental groups, in the time of 1 week, 2 weeks,3 weeks and 4 weeks the four time points, 4 rats were chosen to be sacrificed as the experimental group, rats were killed by overdose of Ether anesthesia and quickiy killed, then separation of the mandible, scraped dig mandibular first molar apical tissue of rats. After fully grinding the organization extracted and purified tissue RNA, it reversed transcribed into cDNA. Real-time PCR was used to detect the expression levels of Btk mRNA of each group. The results were statistically analyzed.Results:1. After operation A group of in 2,3,4 week have increased expression about Btk mRNA were significantly higher compared to the control group, the differences was statistically significant(P<0.05). Btk mRNA expression on 1 week compared to the control group was slightly increased after the operation, the difference was not statistically significant(P> 0.05).2. At 1, 2, 3 and 4 weeks after operation,the expression of Btk mRNA in B group was higher than that in control group, with statistically significant(P<0.05).3. After the operation A, B groups from 1 week to the 2 week Btk mRNA expression levels gradually increased, the expression levels of 2 weeks up to the peak, the expression levels of three weeks began to decrease, the expression levels of four weeks continued to decrease. The expression of Btk mRNA in A group campared with each other at 1, 2, 3 and 4 weeks after operation, the differences was statistically significant(P<0.05).Group B in the expression of Btk mRNA at the 2 week and 3 week campared with other time point,the difference with statistically significant(P<0.05).4. A, B groups at the four points time after operation 1 week, 2 weeks,3 weeks, 4 weeks, group B Btk mRNA expression levels were higher than the A group, with statistically significant(P<0.05).Conclusion:1. Btk involved throughout the development and progression of periapical periodontitis,and in this process the expression of Btk increased.2. Enterococcus faecalis presence are enhanced the expression of Btk in rats apical periodontitis, and its expression closely related to the process of periapical periodontitis.
