Ganoderma lucidum is a precious health-care edible medicinal fungus in China. Ganoderma triterpenoids has a wide range of physiological activities and pharmacological effects, such as reducing blood fat, analgesic, anti-inflammatory, hepatoprotective, enhance immunity, anti-aging, inhibit cancer cell growth and kill tumor cells. However its biological activity in vivo has been greatly affected due to its water insoluble properties.In present study, using Ganoderma triterpenoids as the model drug, three different carriers were studied including bovine serum albumin (BSA) nanoparticles, liposomes, chitosan nanoparticles. The three kinds of nanoparticles were evaluated by the encapsulation efficiency, molecular characterization, stability storage.In this paper, Ganoderma triterpenoids BSA nanoparticles were prepared by desolvation-chemical crosslinking method, Ganoderma triterpenoids nanoliposomes were prepared by the film-ultrasonic method, Ganoderma triterpenoids chitosan nanoparticles were prepared by ionic cross-linking method. The prescription of nanoparticles was optimized by orthogonal design. The optimal preparation crafts of Ganoderma triterpenoids BSA nanoparticles:the quality of BSA was 8mg, the mass of Ganoderma triterpenoids was 8mg, the volume of 0.25% glutaraldehyde solution was 125 μL, the speed of stirring was 400 r/min, the time of crosslinking was 12 h. The optimal preparation crafts of Ganoderma triterpenoids nanoliposomes:the mass proportion between cholesterol and lecithin was 0.150:1, the amount of Ganoderma triterpenoids was 10mg, volume of Tween 80 was 180μL, pH value was 7.0, the hydration temperature was 40℃. The optimal preparation crafts of Ganoderma triterpenoids chitosan nanoparticles:the concentration of chitosan was 2.5mg/ml, the concentration of TPP was 1.5mg/ml, the ratio of acetic acid to ethanol 1:1, the amount of Ganoderma triterpenoids was 4mg.The result of Ganoderma triterpenoids BSA nanoparticles:encapsulation efficiency was maintained to (80.10 ±1.05)%, the drug-loading efficiency was (11.36 ± 0.13)%, the mean diameter was 189 nm. The result of Ganoderma triterpenoids nanoliposomes: the encapsulation efficiency was maintained to (80.36 ± 0.71)%, the drug-loading efficiency was (3.38 ± 0.029)%, the mean diameter was 167 nm. The result of Ganoderma triterpenoids chitosan nanoparticles:the encapsulation efficiency was maintained to (77.05 ± 0.53)%, the drug-loading efficiency was (10.96 ± 0.068)%, the mean diameter was 158 nm.Fourier transform infrared spectroscopy (FT-IR) and Transmission electron microscope (TEM) was used to characterize the structure of Ganoderma triterpenoids nanoparticles. It confirmed that the Ganoderma triterpenoids was successfully encapsulated into the nanoparticle materials. The optimized operation of Ganoderma triterpenoids nanoparticles were reasonable practicable with low particle size with homogeneous microspheres shape. With the particle size, PDI and leakage rate as index, the index in the storage process was not changed greatly, proof of nanoparticles has good storage stability.
