Gut immunity is the major immune defense mechanism against foreign pathogens in Drosophila. To study the effect of the Chinese herbal medicines on oxidative stress-induced gut immunity of Drosophila. Drosophila was treated by paraquat and H2O2 with or without Chinese herbal medicines, and the effect of 50 kinds of Chinese herbal medicine on the survival rate of Drosophila in response to oxidative stress were recorded. And the antioxidant effect of Chinese herbal medicines on lifespan and the number of ISC and enteroblast (EB) in intestinal epithelium cells were analyzed. The extracts from Trichosanthes kirilowii Maxim., Carthamus tinctorius L., Ligusticum chuanxiong Hort. and Chrysanthemum monfolium Ramat. could significantly improve the survival rate of paraquat and H2O2-induced Drosophila, increase the lifespan of Drosophila and regulate paraquat-induced and age-related changes in the number of ISC and EB. These results demonstrate that the extracts from Trichosanthes kirilowii Maxim., Carthamus tinctorius L., Ligusticum chuanxiong Hort. and Chrysanthemum monfolium Ramat. could significantly improve the oxidative stress-induced gut immunity of Drosophila.To study the anti-inflammatory effect of the Chinese herbal medicines. In this study, Drosophila is used as research material. Drosophila was treated by SDS and NaCl with or without Chinese herbal medicines, and the effect of 50 kinds of Chinese herbal medicine on the survival rate of Drosophila in response to anti-inflammatory effect were recorded. And the number of ISC and EB in intestinal epithelium cells, intestinal epithelial cell death, intestinal morphology changes and the expression of antimicrobial peptides were analyzed. Aqueous extracts from Codonopsis pilosula (Franch.) Nannf, Saussurea lappa (Decne.) C.B.Clarke, Imperata cylindrical var. major (Nees) C.E.Hubb. and Melia toosendan Sied.Et Zucc. increased fly survival rate, reduced epithelial cell death, improved gut morphology and promoted the expression of antimicrobial peptide genes. However, we did not observe that these extracts increased SDS (Sodium dodecyl sulfate)-induced cell proliferation in vivo. These results demonstrate that strong anti-inflammatory effects in extracts of Codonopsis pilosula (Franch.) Nannf, Saussurea lappa (Decne.) C.B.Clarke, Imperata cylindrical var. major (Nees) C.E.Hubb. and Melia toosendan Sied.Et Zucc. on Drosophila intestinal cells, and these four kinds of medicine could significantly improve the gut immunity of Drosophila.
