| Objective To observe the effect of Lianpu clearing stomach decoction on the expression of TNF-a(tumor necrosis factor),IL-1β(interleukin 1β),SS(somatostatin)and Gas(gastrin) in serum of chronic gastritis model rats with splenogastric hygropyrexia syndrome established by 2% sodium salicylate lavage and foods with high fat and sugar high quantity of heat, plus the hot and humid environment, in order to provide theoretical support of the treatment mechanism of Lianpu clearing stomach decoction for the treatment of chronic gastritis with splenogastric hygropyrexia syndrome.Methods We got 70 SD rats. Except for 10 normal ones, the other 60 were given 2% of sodium salicylate of 10 ml/(kg·d) by gavage, and fed with high fat and high sugar diet at the same time(20% honey water to drink freely, grease of 10g/kg to take by gavage On alternate days, and 2ml/200 g white wine to take on the days without grease), and they were put in a hot and humid environment(temperature 30℃, humidity 90%) for continuous 20 days, to make chronic gastritis with splenogastric hygropyrexia syndrome. The 60 rats which were made successfully into models were divided into model group, high Lianpu clearing stomach decoction, medium Lianpu clearing stomach decoction group, low Lianpu clearing stomach decoction group, san-jiu weitai group and ranitidine group according to random number table method, and were given the drug by irrigation for 10 days, then were given fasting for 16 h at the end of the last treatment. Afterwards, We used 2% pentobarbital sodium anesthesia by intraperitoneal injection of 0.2ml/100 g, put them in caesarean sectionand then we took the heart blood and finally they were killed by cervical dislocation.Then we took their stomaches quickly besides clipping the amount of gastric antrum in 4% paraformaldehyde.After 24 hours, we took materials and put them on dehydration besides keeping them for backup.After that we use routine HE staining under the microscope to observe the pathologic changes of gastric mucosa and ELISA method to detecte the content change of TNF-a(tumor necrosis factor), IL-1 β(interleukin 1 β), SS(somatostatin)and Gas(gastrin) in the rats.Results ①The general situation of rats Normal group rats weight were increased day by day, and they responsed flexiblely, food intake and water amount is no exception. Defecate is normal, and their hair is lucidus. Model group: In the first week after building,weight gain relatively obvious, the late periods becom stable gradually.9 days after building,the water quantity is reduced gradually, besides food intake.Their stool is thin pond, hair is less lucidus, and their respond is stubborn.As for Lavage dosage groups,after about a week the situation of the stool.hair.food intake and water amount had been increased.Even the low-dose group of Lianpu Clearing stomach decoction generally improved,but compared to the other groups,the effect is not obvious.The most obvious improvement happened in the medium and high groups. ② Rat gastric mucosa pathological tissue section:Four layer structure of norm group is clear, complete,and there is no obvious abnormalities.In model group rats mucosa there is a large number of columnar epithelium cell falling off, incomplete; Gastric pit are visible in more loss of epithelial cells, gastric and visible part is broken, and a few wall cell is swelling.There is a circular transparent halo,the cytoplasm staining is shallow. Little neutrophilic infiltrate was also visible in the lamina propria.Lianpu Clearing stomach decoction high, medium dose group, san-jiu weitai group and ranitidine group rats stomach epithelial cells are relatively complete, the glands were arranged in order.A small amount of exfoliated cells within the glandular lumens are known as fragments, low dose group of epithelial shedding is clear compared to the other group which is less than model group, of which Lianpu Clearing stomach decoction medium group and the high group is most obvious. ③TNF-a(tumor necrosis factor), IL-1β(interleukin 1 β)in model group were higher than those in normal group and the TNF-a(tumor necrosis factor), IL-1β(interleukin 1 β)of Lianpu Clearing stomach high group, and medium group were lower than other drug group.④ SS group were lower than normal group, the SS of Lianpu Clearing stomach low group,medium group,high group and ranitidine group, san-jiu weitai group were increased after corresponding drug treatment which were higher than that of model group but were lower than the normal group The SS of Lianpu Clearing stomach medium group and high group were higher than other drug groups.Conclusion ① Animal models using multi factors of inflammatory factors and fatty and sweet diet and external hot and humid environment were very similar with the lesion characteristics of clinical chronic gastritis of spleen stomach damp heat. The method combines multi internal and external factors to manufacture model that is same to the cause of clinical chronic gastritis of spleen stomach damp heat. Besides the method is simple with good reproducibility.②By inhibiting the expression of proinflammatory cytokines of TNF-a(tumor necrosis factor), IL-1β(interleukin 1 β), Lianpu Clearing stomach decoction was able to make a variety of effects such as stopping all kinds of stimulation to produce excessive immune responses causing tissue injury, preventing producing a variety of inflammatory mediators, reducing epithelial cell hyperplasia, repairing damaged tissue and promoting the absorption of inflammation at the same time reducing the exudation of inflammatory.Besides,it can reduce coagulation in Microcirculation and promote the circulation of local blood thus improving tissue nutrition.③By enhancing the expression of somatostatin and reducing the secretion of gastrin, Lianpu Clearing stomach decoction can protect gastric mucosa and promote the absorption of inflammation.In addition, Lianpu Clearing stomach decoction play a good therapeutic and improvement effect in Chronic gastritis with splenogastric hygropyrexia syndrome.④Compound Chinese medicine composition is very complex, We need to do further study from multi angle on its pharmacological action. However, due to the experimental conditions.funds and so on, this study only chooses some indexes to do preliminary exploration about chronic gastritis. Therefore, the shortcomings need to be strengthened and improved. |
