Background and Objiective: Colorectal cancer is one of the most common malignancies. In recent years, the incidence and deathrate of colorectal cancer is increasing; invasion and metastasis are the major cause of treatment failure and death in patients with colorectal cancer. It is urgent to find out the invasion and metastasis associated factors, so as to reveal the essence of the invasion and metastasis, and it will have great significance for exploring new means of anti-tumor. TMPRSS4 is one of TTSPs, E-cad is a member of Ca2+dePendent cell adhesion molecules family,Study indicated that TMPRSS4 and E-cad plays an important role in tumor adhesion, invasion and the process of development. In this study,we detected the expressions of MPRSS4 and E-cad in colorectal cancer by immunohistochemical(SP method), analyzed the relationship of MPRSS4 and E-cad, and investigated their regulating in the development of colorectal carcinoma, it might provide new means of diagnosis prognosis evaluation, gene therapy and immunotherapy for colorectal carcinoma.Materials and Methods: 60 cases were obtained from surgical Specimens of patients with colorectal carcinoma hospitalized in the department of general surgery of the first hospital affiliated to Henan University from January in 2008 to December in 2009, all the patients diagnosed to be colorectal carcinoma by pathology;all the patients had not received preoperative radiotherapy or chemotherapy. 20 cases of normal adjacent mucosa and 20 cases of colorectal adnoma served as normal control groups, the expressions of TMPRSS4 and E-cad protein were detected by immunohistochemistry(SP method), the percentage of positive cell represented positive rank, we analyzed the datas by SPSS17.0, the statistical methods included McNemar test and Chi-square pearson contingency coefficient.The clinical significance was carried out by way of combining with the clinicopathological indexes and survival.Results:1.The positive rate of TMPRSS4 in colorectal cancer, colorectal adnoma and normal adjacent mucosa was 60.0%、20.0%、10.0%, respectively. TMPRSS4 in colorectal cancer was significantly higher than that in normal adjacent mucosa(P=0.000),and colorectal adnoma(P=0.002). there was no statistical significance between the expression of normal adjacent mucosa and colorectal adnoma(P>0.05), In colorectal cancer, the expression of TMPRSS4 has no relation with some clinical parameters; including sex(P=0.832),age(P=0.576), tumor location(P=0832,tumor size(P=0.825), gross morphology(P=0.574), Pathological type(P=0.623),and tumor differentiation(P>0.05); But it had great relationships with the invasive depth, lymph node metastasis, Dukes stages(P=0.908),2. The Positive rate of E-cad in colorectal cancer, colorectal adnoma and normal adjacent mucosa was 25.0%、50.0%、70.0%; respectively, E-cad in colorectal cancer was significantly lower than that in normal adjacent mucosa(P=0.000), and colorectal adnoma(P=0.037),but there was no statistical significance between normal adjacent mucosa and colorectal adnoma. In colorectal cancer, the expression of E-cad had great relationshiPs with the invasive depth(P=0.015), lymph node metastasis(P=0.035), Dukes stages(P=0.025), but has no relations with some clinical parameters including age(P=0.527), sex(P=0.880), tumor size(P=0.086), tumor location(P=0.764), gross morphology(P=0.690)and pathological type(P=0.096); 3. In colorectal cancer,the expression of TMPRSS4 and E-cad were differently(χ2=9.76,P<0.05); the expression of TMPRSS4 and E-cad was related(P<0.05,r=-0.607)Conclusion: 1. Both TMPRSS4 and E-cad protein were related with colorectal carcinoma’s genesis. 2. The high expression of TMPRSS4 protein and low expression of E-cad protein prognosticate the stronger ability of colorectal cancer to invase and metastasize. 3. TMPRSS4 and E-cad were negative correlation in colorectal cancer, and detecting the expressions of TMPRSS4 and E-cad protein will be good for diagnosis and judgement of prognosis. It also provide new means of immunotherapy and gene therapy for colorectal cancer.
