The Research On The Role Of Human Lung Cancer A549 Cell Of PTX-loaded PH-sensitive Polymeric Micelles

Objective: To study the effect of taxol drug carrier system human lung cancer A549 cells and the action mechanism of it, to provid a basis for the clinical test.Method: This experiment used A549 cells as experimental object, free screening suitable conce ntration of paclitaxel as positive control drug, to obtain the objective of the experiment.MTT meth od of blank material was used to detect whether the drug carrier have the toxicity to human lung cancer cells of A549 cells; MTT method to check the effect of the drug loading paclitaxel to thi s tumor cell; Applying the RT-PCR skill to check the expressions of Bcl-2 and Bax mRNA and t he Western blot skill to check the expressions of Bcl-2 and Bax protein; Kit method to detect cell supernatant of LDH and cell content of SOD and MDA.Result: The Blank material of taxol drug carrier system had no effect on cells; Drug taxol co uld arrest the cell cycles, and put up the concentration dependence; the Hoest33342/PI result make clear that when the drug content increased, the degree of cell apoptosis and necrosis became grea ter, the quantity of Bcl-2 was inhibited and the Bax was promoted, the culture supernatant of the cell lines has low LDH content, and in the cell,MDA content was increased and the SOD conten t was reduced.Conclusion: drug taxol had an inhibitory effect to the human lung cancer A549 cells.
