This dissertation is composed of four chapters. The first chapter study the mechanism, test indexes and the principle of coagulation. The second chapter study the influence of Paeonia lactiflora Pall. extract and compounds on the coagulation time. The third chapter explores the effect of Paeonia lactiflora Pall. extract and compounds of fibrinolytic activity and fibrinolytic activity of urokinase defibrase with fibrin plate method. The fourth chapter summarized natural drugs on blood coagulation function recently.Chapter 1. Introduction The mechanism, test indexes and the principle of coagulation.Chapter 2. Influence of Paeonia lactiflora Pall. extract and compounds on the coagulation time The plasma recalcification time(PRT), prothrombin time(PT), activated partial thromboplastin time(APTT), thrombin time(TT), fibrinogen(FIB) were used to assay effect of Paeonia lactiflora Pall. extract and compounds on the blood coagulation. Results showed that Paeonia lactiflora Pall. extract and compounds can prolong PRT, APTT and PT. The effects of ethyl acetate extract of PRT than that of breviscapine(P<0.001) as the positive control. There was no significant difference on APTT between the effects of n-butanol extract and breviscapine(P>0.05). Compared with breviscapine, n-butanol, ethyl acetate and petroleum ether extract could significantly prolong the PT(P<0.001). Paeonia lactiflora Pall. can prolong the time in TT assay,but the effect is far less than breviscapine. Ethyl acetate extract could significantly increase the content of FIB, but the compounds isolated in Paeonia lactiflora Pall. reduced FIB content.Chapter 3. Fibrinolytic activity and effect on urokinase fibrinolytic activity of Paeonia lactiflora Pall. extract and compounds Fibrin plate method was used to evaluate the dissolution of Paeonia lactiflora Pall. extract and compounds on the fiber protein and influence on fibrinolytic activity of urokinase. The results showed that Paeonia lactiflora Pall. extract and compounds had no fibrinolytic activity, but on urokinase fibrinolytic activity inhibition, where in ethyl acetate extract(IC50=0.11 mg/m L), ethanol extract(IC50=0.63 mg/m L), petroleum ether extract(IC50=1.54 mg/m L), n-butanol extract(IC50=0.16 mg/m L), pentagalloyglucose(IC50=0.02 mg/m L) on urokinase fibrinolytic activity of the strongest inhibition. The effects are better than that of compound danshen tablets(IC50=6.28 mg/m L) as the positive control.Chapter 4. Effect of natural drugs on coagulation effect Natural drugs anticoagulant and procoagulant effect are summarized.
