Objective:To discuss the influence on nitric oxide (NO), cyclic guanosine monophosphate (cGMP) of SD rats with renal ischemia reperfusion injury (RIRI) by different intervention schemes of curcumin, to explore the protective effect and mechanism of curcumin on RIRI.Methods:60 SD rats were randomly divided into five groups,12 rats per group, and they were all given RIRI model preparation by unilateral artery occlusion method. Rats in Sham group were not given occlusion of left renal artery,and rats in Model group were given full cirulation after occlusion of left renal pedicle for 45 min. Based on Model group, Rats in CUR group were given curcumin by 100mg/kg 2h before occlusion of left renal pedicle. Rats in IPO group were given curcumin by 100mg/kg on 45 min later after occlusion of left renal pedicle. Rats in CUR+IPO group were given curcumin by 100mg/kg 2h before occlusion of left renal pedicle, and given curcumin by 100mg/kg 30min later after reperfusion. All the rats were continuely breed by metabolic cages for 24h after intervention, and β2-MG in urine in all groups were compared, and UAER were calculated. Inferior vena cava blood was extracted 24h after reperfusion, then they were all killed and left kidneys were resected. Serum Cr and BUN were compared. Paraffin sections of renal tissue were made to observe tissue morphology, also tubular scores were evaluated by Paller score standard, and kidney homogenate were made to test content or activity of NO, NOS, cGMP.Results:β2-MG and UAER in urine, serum Cr and BUN, content or activity of NO, NOS and cGMP in renal tissue homogenate in group of Model, CUR, IPO, CUR+IPO were all higher than those in Sham group(P<0.05 or P<0.01), and above index in group of CUR, IPO, CUR+IPO were all lower than those in Model group(P<0.05 or P<0.01), yet there were no statistical difference on above index between CUR group and IPO group(P>0.05), and above index in CUR+IPO group were all higher than those in CUR group and IPO group(P<0.05), and the renal pathological findings also supported the above results.Conclusions:(1) It can copy RIRI animal model by rats by unilateral artery occlusion method of excision one kidney and occlusion of contralateral renal artery. (2)It can effectively play the protective role of RIRI by intervention of curcumin, and reduces renal structural damage, increases kidney tissue activity, improves kidney function, and it may be associates with the activation of NO and cGMP. (3)It has better synergistic effect on reducing RIRI by CUR+IPO intervention schemes of curcumin, and has better protective effect on kidney.
