Objective:Through the establishment of cerebral ischemia reperfusion model in rats,He ad hole through thorn on migration of neural stem cell transplantation after cerebral ische mia in rats.Methods:This experiment adopts the improved line bolt of middle cerebral artery cere bral ischemia reperfusion model(MCAO) and rats were randomly divided into:control grou p, model group,transplantation group,the head of needle group.In NSCs transplantation 24 h after intervention measures,can be the hole through the temples,twisting line technique for2 minutes,once per day,per treatment time of 15 min.In 3days,7days,14 days,21days of each group rats were observed:using sevenpoint scale assessment standard assessment nerve disfu nction conditions;TCC staining observation the change of the infarct size;BDNF,bFGF imm unohistochemical metho d to observe the correlation;Brdu/GFAP,Brdu/NSE fluorescence dou ble standard method to observe the ischemic area of positive cells expression;Statistical cor relation area number of positive cells.Results:1.each group rats nerve function defect symptom scores declined,head of ne-edle grou p compared with other groups of each point in time,reduce the most significantly,the differ ence was statistically significant(P<0.05);2.the first needle group BrdU positive cells begin to increase when 3d,14 d reached th e peak,when compared with transplants 14 d group,number of positive cells increased signifi cantly(P<0.05);3.the first needle group of Nestin positive immune cells 3d began to increase,14 d pea k,pin 7d group compared with the transplant 7d group,number of Nestin positive cells incr eased significantly(P<0.05);4.after cerebral ischemia injury BrdU/NSE,BrdU/GFAP positive cells increased,the hea d of the needle group 14 days BrdU/NSE,BrdU/GFAP positive cells expression peak,compar ed to transplantation group,model group, with significant difference(P<0.05);5.the first needle group 21 days the expression of BDNF positive cells peak,compared with 21 days transplantation group had significant difference(P<0.05);6.Pin 7days group bFGF immune positive cells expression to the peak,compared with transplantation group,model group,there are significant differences(P<0.05).Conclusions:1.The head acupuncture point through thorn therapy could significantly promote the NSCs transplantation in rats after cerebral ischemia rats balance,the grasping ability,promotes neuralfunctional recovery;2.The head acupuncture point through thorn can promote the transplantation of NSCs into neurons and glial cells differentiation,and can promote the transplantation of NSCs proliferation and the expression of neurotrophic factor BDNF and bFGF;3.The head acupuncture point through thorn by promoting proliferation differentiation of neural stem cells after transplantation and the expression of neurotrophic factor BDNF,bFGF,thus enhanced the NSCs effect for the treatment of cerebral ischemia.
