[Objective]In order to text the TNF-A-308, IL-1B+3953 and MMP-2-1306 gene polymorphism. The gingival cells obtained from CP/AgP patients who were from department of periodontics of Affiliated Stomatology Hospital Of Kunming Medical University. And the volunteers group. This study was to investigate the correlation between IL-1B+3953, MMP-2-1306 and TNF-A-308 gene polymorphism and the susceptibility and the extent of damage of periodontitis.[Methods] 101 patients with periodontitis Including 80 CP patients and 21AgP patients.23 volunteers served as the control group. According to the severtity of, CP group is divided into slight, moderate and severe.After the informed consent of the patients, the exfoliative cells was obtained from the oral mucosa swab. IL-1B+3953, MMP-2-1306 and TNF-A-308 which were detected by PCR-RFLP method, after DNA were extracted by Chelex-100 method and nested PCR amplification. The datas were analyzed by Haplo view 4.2 software after genotype distribution was identified to accord to the Hardy Weinberg (Hardy-Weinberg, HW) balance. The software was utilized to analyze the distribution differences between chi-square and risk degree of logistic regession of the care group and the control group in a singe SNP loci of genotye and allele.The online tools SHEsis (http://analysis. bio-x.cn/SHEsisMain.htm) was utilized to check the repeatability of the data. Statistical differences (P<0.05), with relative risk degree OR>1 risk increases, the OR<1 lower risk.[Result] 1. Each genotype of MMP-2-1306, in CT,CC and TT was observed in the periodontitis group (slight, moderate and severe CP and AgP) and the control group The frequent distribution of T and C in the periodontitis group allele by chi-square test and logistic regression analysis have no difference in the correction of age and gender(P>0.05).2. Each genotypes of IL-1B+3953 in CT,CC and TT was observed in the periodontitis group (slight, moderate and severe CP and AgP) and the control group The frequent distribution of T and C in the periodontitis group allele by chi-square test and logistic regression analysis had no difference in the correction of age and gender(P>0.05).3. The frequency of distribution of genotypes AA,AG and GG and the control group of AgP group TNF-A-308 by chi-square test and logistic regression analysis, The frequency distribution of A and G allele in the AgP group, by chi-square test and logistic regression analysis, adjusted for gender and age were not statistically significant(P>0.05).4. The frequency of distribution of genotypes AA,AG and GG and the control group of moderate CP group TNF-A-308 tested by chi-square test and logistic regression analysis, adjusted for gender and age were not statistically significant(P>0.05); The frequency distribution of A and G allele in the moderate CP group, by chi-square test and logistic regression analysis, adjusted for gender and age were not statistically significant(P>0.05).5. The difference was statistically significant in terms of slight CP group TNF-A-308 A and G allele(P=0.011) compared with control, logistic regression analysis showed that the TNF-A-308 mutant A alleles rise the risk of slight CP besides, TNF-A-308 wild type G allele reduced the risk of slight CP, which it was a protective factor.6. Severe CP group of TNF-A-308 after adjusting for gender and age showed that TNF-A-308 genotype GG had significant differences in the severe CP group and the control group, which can be seen as the protective factors; The frequent distribution of A and G allele after correction of gender and age showed the TNF-A-308 mutant A allele in severe CP groups increased significantly, logistic regression analysis showed that the TNF-A-308 mutant A alleles rise the risk of severe CP. Besides, TNF-A-308 wild type G allele reduced the risk of severe CP, which it was a protective factor.[Conclusion]1. IL-1B+3953 gene polymorphism might not be associated with susceptibility to CP and AgP.2. MMP-2-1306 gene polymorphism might not be related to the CP and AgP susceptibility is no relevant.3. The TNF-A-308 allele of A may cause the risk of suffering from moderate CP increased significantly, G allele with moderate CP risk decreased significantly.4. The TNF-A-308 allele of A may cause the risk of suffering from severe CP increased significantly and G allele with the risk of severe CP was significantly decreased.5. There was no correlation between TNF-A-308 gene polymorphism and the AgP.
