Obj ective:To Study the effects of antibacterial peptide LL-37 on bladder cancer T24,EJ cells andimmortalizedhumanurothelial cell line SV-HUC-1, and to explore its possible mechanism.Methods:1.The bladder cancer T24, EJ cellsand immortalized human urothelial cell line SV-HUC-1 were treated With different concentrations of antibacterial peptide LL-37,MTT cell proliferation assay was used to observe the inhibitory effect.,and use the microscope to observe the cell morphology.2.The bladder cancer T24, EJ cellsand immortalized human urothelial cell line SV-HUC-1 were treated With different concentrations of antibacterial peptide LL-37,and detect thecell apoptosis by FCM.3.The bladder cancer T24, EJ cells were treated With different concentrations of antibacterial peptide LL-37,and detect the cell cycle by FCM.4.The bladder cancer T24, EJ cells were treated With different concentrations of antibacterial peptideLL-37,use the Western blot to detect the changes of Bax, Bcl-2 protein expression.Results:1.The antibacterial peptide LL-37 inhibited the proliferation of bladder cancer T24, EJ cells in a dose-dependent manner;it has no obviously effect to immortalized human urinary tract epithelial SV-HUC-1 cells.2.The bladder cancer T24, EJ cells was treated with antibacterial peptide LL-37 after 24hours, cellswere increased in S-phase, and it increased with the concentrations of antibacterial peptide LL-37.3.The bladder cancer T24, EJ cells was treated with antibacterial peptide LL-37 after 24hours, the expression of Bax was more than that in control group, and it increased with the concentrations of LL-37,and the relative expression increased with the increase of concentrations; The expression of Bcl-2had no obvious changes compared with the control group.Conclusions:1.Antibacterial peptide LL-37 inhibits the growth ofbladder cancer T24,EJ cells, which showed a dose-dependent manner;the antibacterial peptide LL-37 have no obviously effect onhuman urinary tract epithelial SV-HUC-1 cells.2.The antibacterial peptide LL-37 could change the cell cycle,make the cell arrest in S-phase and inhibit the proliferation of bladder cancerT24 and EJ cells.3.Antimicrobial peptide LL-37 couldmake high expression of Bax to inhibit the growth of bladder cancer T24,EJ cells.
