Part oneObjective:To study the CNP expression in patients of Diabetic lower extremity atherosclerosis; and the correlation with lower extremity arteriosclerosis degree.Methods:100 cases of adult diabetic mellitus were selected, except hypertension, including 50 patients with diabetic lower extremity arteriosclerosis and 50 without lower extremity arteriosclerosis. The diagnosis of peripheral atherosclerosis detection based on color Doppler. If lower extremity arterial intimal thickening or plaques appear with the color Doppler examination, diabetic lower extremity atherosclerosis will be determined.5ml plasma was collected from fasting subjects. Using the ELISA method to detect C-type natriuretic peptide plasma sodium.Compare diabetic lower limb arteriosclerosis and diabetes difference C-type natriuretic peptide levels in patients with lower limb arteriosclerosis occurred.T test was used for data processing. Detecting atherosclerosis in patients with diabetic lower extremity limb ischemia stage and ratings, the femoral arterial intima-media,endothelial function (flow mediated brachial artery diastolic),lower extremity arterial stenosis/stenosis at the upper end of 1-2cm normal systolic peak velocity caliber arteries (peak flow ratio), HbAlC,LDL-C,creatinine,duration of diabetes, and compared with the lower extremity diabetic plasma C-type natriuretic peptide levels in patients. With the correlation analysis.Statistical methods for the independent samples T-test.Result:Plasma of Patients with diabetic lower limb arteriosclerosis CNP mean 2.998 ± 2.841pg/ml,Mean plasma CNP content did not occur in patients with diabetic lower limb arteriosclerosis was:5.667 + 1.215pg/ml.The difference was statistically significant. Conclusions:CNP plasma in patients with diabetic lower limb arteriosclerosis content CNP plasma content compared with patients with diabetic lower limb arteriosclerosis occurred significantly lower. The vascular endothelium was injured in the patients with the diabetic lower extremity atherosclerotic, thickening.Vascular smooth muscle proliferation.Thinning the blood vessel lumen diameter.Decreased blood vessel elasticity. The more serious the more arterioles, vascular lumen damage.CNP dilation of blood vessels function is reduced. Thus resulting mainly in small arteries and arterioles supplying severe distal limb ischemia, or necrosis. significant. Conclusions:CNP plasma in patients with diabetic lower limb arteriosclerosis content CNP plasma content compared with patients with diabetic lower limb arteriosclerosis occurred significantly lower. The recombinant vector pCAGGS/mCNP transfected into human umbilical vein endothelial cell line (HUVECs).By pronuclear microinjection to load linearized target gene, followed by transplantation of fertilized eggs in the fallopian tube.ldentification of rat first built by PCR method of expression mCNP.We will be screened out of F1 mice inbreeding,F2、F3。Result:The CNP transgenic mice has been reconstructed.
