Objective:The purpose of this study was to establish a hepatic cell line with HBV genotype I, which could be used in evaluating the effect of drug test on HBV/I. This study determined the distribution characteristics of HBV genotypes and subtypes in countries neighboring China. The progress made in genomic research involving HBV was also reviewed.Method:1) Constructing the eukaryotic expression vectors of 1.2-genome length HBV with three genotypes and transfected to HepG2 cell line.2) Detected the effect of four natural compounds on HepG2 cell line and HBV expression.3) Nine hundred fifty-one complete genome sequences of HBV from 14 countries neighboring China were selected from NCBI. The sequence-related information was analyzed and recorded. All of the HBV genotype B were screened for alignment.Results:1) HBV three genotypes were transient transfected into HepG2 cell line and could expression of surface antigen.2) Over 6000U/mL Interferon alb significantly inhibit the HBsAg expression of Y132 with HBV-I1/C1. HepG2 cell line only sensitive with the compound D.3)The dominant HBV genotypes in the countries neighboring China were genotypes B, C, and D. And the dominant subtypes were adw2 and adrq+. The relationship between genotype and serotype of HBV in these countries was shown to differ from previous research results. As shown by sequence alignment, the sequence divergence between five sub-genotypes (B3, B5, B7, B8, and B9) was< 4%. The B sub-genotypes share six common specific amino acid sites in the S region.Conclusion:This study successfully constructed three eukaryotic expression vectors with 1.2-genome length of HBV. High concentration of alb have inhibitory, effect on recombinant I1/C1. For H75-B3,compound A and D group have a degree of anti-viral activity in 6h.For Y17-C1, compound A,B and D group have a degree of anti-viral activity.The B3, B5, B7, B8, and B9 sub-genotypes can be clustered into quasi-sub-genotype B3.
