ObjectiveNowadays, breast cancer attacks women most among all malignancies and threat-ens their health badly. As one kind of gene which can conduct and transcribe signals, STAT3 regulates and controls a series of physiological activities such as growth, ap-preciation, differentiation and apoptosis in normal cells. But persistent activate of STAT3 would make tumor cells’ability of appreciationand anti-apoptosis go down, make tumor cells transcription affect and transfer, form blood vessels and immune escape, which makes it recognized oncogene at present. By detecting expressionof STAT3 in breast tissues near cancer and in breast cancer tissues, this research analysis the dependency between STAT3 and breast cancer, and explores application prospect of STAT3 in the treatment of breast cancer.MethodCollect 52 primary breast cancer patients who meet a criterion of being brought into and have come to the Third Affiliated Hospital of Kunming Medical University between September 2013 and February 2014. Collect 20 normal breasttissues for comparing. Detect expression level of STAT3 gene in 52 breast cancers and 20 tissues near cancer using immunohistochemical technology. Analysis its expression level in normal breast tissues and breast cancers, in different molecular subtyping(LuminalA, Luminal B, Her-2 over-expression and Basal-like), in different clinical parameters (patients’ age, size of tumors, condition of lymph gland transferand clinical stages and in biological indicators(ER, PR, Her-2)expression rate and dependency..Results1.The STAT3 gene in normal breast tissue and its statistical significance in nor-mal breasttissueThe STAT3 geneexpress 16 in 20normal breast tissues, expression rate is80%. The STAT3 genepositiveexpress 47 in 50 breast cancer tissues,positive expression rate is 90%. The positive expression rate in breast cancer tissues is higher than which is in comparing group. The x2is 0.633 by checkout and compare, P is 0.426>0.005. Still there is no statistical significance between the expression rate in normal breast tissue and breast cancer tissue. Put the expression of STAT3 genein normal breast tissue and breast cancer into four types "-" "+" "++" "+++",the expression of STAT3 gene in normal breast tissue and breast cancer comparingx2is1.966,fisherP=0.005,P=0.007,P <0.05. This shows prominent statistical significance,which is that the higher level of STAT3 genepositive expression rate, the higher risk of contracting breast cancer.2. The dependency between STAT3 gene and four molecular subtypingIn four molecular subtyping of breast cancer molecule, which is LuminalA, Lu-minal B, Her-2 over-expression and Basal-like. There are 11 LuminalA,9 of them are positive expression, and expression rate is 81%. There are 17LuminalB,15 of them are positive expression, and expression rate is 88%.There are 11 Her-2,10 of them are positive expression, and expression rate is 90%.There are 13 Basal-like,13 of them are positive expression, and expression rate is 100%. The positive expression rate in-four molecular subtypingby checkout and comparefisherP=0.514,X2= 2.406, P=0.493 , fisherP>0.05 and there is no statistical significance in difference.Put the expression of STAT3 gene in four molecular subtyping into four types "-" "+" "++" "+++", the expression of STAT3 gene in four molecular subtyping comparingX2is 3.925, fish-erP=0.943,P=0.0916,P>0.05 and there is no statistical significance in difference, whichis thatthe positive expression rate difference of STAT3 gene in different mo-lecular subtyping (LuminalA, Luminal B, Her-2 over-expression and Basal-like) shows no statistical significance (P>0.05) and no dependency.3. The dependency between STAT3 gene andbreast cancerbiological indicator3.1 There are32 ER positive in the breast cancer group,30 of them are positive expression in STAT3 gene, and expression rate is 81%. There are 20 ER negative,17 of them are positive expression in STAT3 gene, and expression rate is 93%.Put the expression of STAT3 gene in ER grouping into four types "-" "+" "++" "+++",the ex-pression of STAT3 genecomparingx2=4.370,fisherP=0.255,P=0.224,P>0.05 and there is no statistical significance in difference,whichis thatthere is no dependency be-tweenSTAT3 gene and ER.3.2 There are29PR positive in the breast cancer group,25 of them are positive expression in STAT3 gene, and expression rate is 86%.There are 23 PR negative,22 of them are positive expression in STAT3 gene, and expression rate is 95%.Put the expression of STAT3 gene in PR grouping into four types "-" "+" "++" "+++",the ex-pression of STAT3 genecomparingX2=3.050,fisherP=0.454,P=0.384,P>0.05and there is no statistical significance in difference,whichis thatthere is no dependency between STAT3 gene and PR.3.3 There are17Her-2positive in the breast cancer group,14 of them are positive expression in STAT3 gene, and expression rate is 82%. There are 35 Her-2 negative, 31 of them are positive expression in STAT3 gene, and expression rate is 88%. Put the expression of STAT3 gene in Her-2 grouping into four types "-" "+" "++" "+++", the expression of STAT3 genecomparingx2=2.386,fisherP=0.541,P=0.496,P>0.05 and there is no statistical significance in difference,whichis thatthere is no dependency between STAT3 gene and Her-2.4. The dependency between STAT3 gene and different breast cancer clinical fea-tures4.1 In the group which takes age as measure, there are 12age-under-50 in the breast cancer group,9 of them are positive expression in STAT3 gene, and expression rate is 75%. There are 40 age-above-50,38 of them are positive expression in STAT3 gene, and expression rate is 95%. Put the expression of STAT3 gene in age grouping into four types "-" "+" "++" "+++", the expression of STAT3 gene comparing x2=6.621,fisherP=0.084,P=0.085,P>0.05 and there is no statistical significance in dif-ference,which is that there is no dependency between STAT3 gene and age. Put the expression of STAT3 gene into two types "-" "+",the expression of STAT3 gene comparing X 2=2.259,P=0.133,P>0.05 and there is no statistical significance in differ-ence.4.2 In the group which takes size of tumor as measure, there are5size-under-2cm in the breast cancer group,3 of them are positive expression in STAT3 gene, and ex-pression rate is 60%. There are 47size-between-2cm-and-5cm,44 of them are positive expression in STAT3 gene, and expression rate is 93%. Put the expression of STAT3 gene in sizeof tumorgrouping into four types "-" "+" "++" "+++", the expression of STAT3 genecomparingX2=6.650,fisherP=0.128,P=0.084,P>0.05 and there is no statis-tical significance in difference,whichis thatthere is no dependency between STAT3 gene and size of tumor. Put the expression of STAT3 gene into two types "-" "+",the expression of STAT3 gene comparingX2=2.645,P=0.104,P>0.05 and there is no sta-tistical significance in difference.4.3 In the group which takes lymph gland transfer or not as measure, there are231ymph-gland-transfer in the breast cancer group,23 of them are positive expres-sion in STAT3 gene, and expression rate is 100%. There are 291ymph-gland-do-not-transfer,24 of them are positive expression in STAT3 gene, and expression rate is 82%. Put the expression of STAT3 gene in lymph gland transfer or not group-ing into four types "-" "+" "++" "+++", the expression of STAT3 genecomparing X2=4.999,fisherP=0.212,P=0.834,P>0.05 and there is no statistical significance in dif-ference. But Put the expression of STAT3 gene in lymph gland transfer or not group-ing into two types "-" "+",the expression of STAT3 genecomparing X2=4.387, fish erP=0.046,P=0.036,P<0.05and there is statistical significance in difference, which is that the higher level of STAT3 genepositive expression rate, the higher risk of lymph gland transfer.4.4 In the group which takes clinical stages as measure,there are7 I Ain the breast cancer group,6 of them are positive expression in STAT3 gene, and expression rate is 85%. There are 12 I B in the breast cancer group,10 of them are positive ex-pression in STAT3 gene, and expression rate is 83%.There are21ⅡA in the breast cancer group,19 of them are positive expression in STAT3 gene, and expression rate is 90%.There are12ⅡBin the breast cancer group,12 of them are positive expression in STAT3 gene, and expression rate is 100%.Put the expression of STAT3 gene in clinical stages grouping into four types "-" "+" "++" "+++", the expression of STAT3 genecomparingx2=4.567,fisherP=0.848,P=0.204,P>0.05 and there is no statistical significance in difference,whichis thatthere is no dependency between STAT3 gene and size of clinical stages. Put the expression of STAT3 gene into two types "-" "+" ,the expression of STAT3 gene comparing x2=0.058,P=0.971,P>0.05 and there is no statistical significance in difference.Conclusion1.STAT3 gene shows 90% of positive expression rate in 52 cases of breast cancer tissues.2. The positive expression ratedifference of STAT3 gene between breast tissues near cancer andbreast cancer tissuesshows prominent statistical significance, which is that the higher level of STAT3 genepositive expression rate, the higher risk of con-tracting breast cancer.3. The positive expression rateof STAT3 gene in four molecular subtyping (Lu-minalA, Luminal B, Her-2 over-expression and Basal-like) shows no statistical sig-nificance.4. The positive expression rate of STAT3 gene in different biological indicators (ER, PR, Her-2) shows no statistical significance.5. There is no statistical significance between the positive expression rate of STAT3 gene and patients’ age, size of tumors or clinical stages (P>0.05). But these is statistical significance between the positive expression rate of STAT3 gene and condi-tion of lymph gland transfer. The higher level of STAT3 genepositive expression rate, the higher risk of lymph gland transfer.
