Objective In this experiment, the skin of rabbits was irradiated by IPL device, the epidermis thickness and dermal thickness were measured then compared the calculation of collagen fiber area.. A preliminary study on the effect of IPL removal effect on rabbit skin and topical Ketoprofen Gel on skin.Method1.Ten health rabbits with exposed back skin and divided to a,b,c,d four groups in the back.Group a is IPL therapy area,group b is IPL combination with ketoprofen gel area,group c is ketoprofen therapy area and group d is control area.The area of each group is 2cm×2cm.2.IPL source is EPOSE-Y type, the wave length from620nm~1200nm,the pulse duration is 20 ms,first pulse is 7ms,second pulse durationis 12 ms,the fluence is 28J/cm2 and the sport size is 40mm×8mm.After treatment use ketoprofen gel once perday. Skin biopsy was respectively taken from rabbits 7d,15 d,28d after treatment from each group of rabbits skin.The succeeded and typical specimens were selected 10%formalin-fixed, paraffin-embedded, histological sections(5μm thick)were cut consecutively,stained by.hematoxylin-eosin(H.E) and van gieson’s.3.The pathologic transformation past-treatment was obserbed under light microscope,and to measurement thickness of epiderms,dermis and to detect the change of collagen fiber content in the dermis by Image-pro puls 6.0 system image analyses Software.4.statistical methods:the quantitative data were shown by means plus or substractiong standard deviation,carry through homogenetity of variace one-way ANOVA.Results1. Observed the thickness of the epidermis under the microscope.IPL treatment group: after 7d,the IPL treatment group was obserbed under the light microscope, the structure of the epidermis was complete, the cell layer was clear, and the degeneration and necrosis of the basal cells was partly destroy.IPL+ ketoprofen gel in the treatment group: irradiation after 7d epidermal layers structure, clear and complete, hierarchy and the basement cell layer degeneration and necrosis are clear.compared to the control group, the difference is statistically significant(P < 0.05).2, Observation the thickness of the dermis under the microscope IPL irradiation group: after7 d irradiation, the papillary congestion was congested,and the infiltration of lymphocytes was seen.The nucleus of the inner root sheath of the hair follicles is fixed, the cytoplasm is vacuole, the hair disappears and the circular cavity is left. Can also see unremoved hair follicles.IPL+ ketoprofen gel group: irradiation after 7d dermal papilla layer with mild edema and some inflammatory cell membrane shrinkage, rupture, nuclear fragmentation and pyknosis that reduce the infiltration of inflammatory cells.7d after irradiation, IPL irradiation group and IPL+ ketoprofen gel in the treatment group compared to the dermal thickness have significant difference(P < 0.05) and 28 d after irradiation, increased group dermal thickness of the IPL treatment group, IPL+ketoprofen gel treatment, with significant difference(P < 0.05).3.collagen fiber area ratio IPL irradiation group: after irradiation, the collagen fiber appeared broken, the morphology of the collagen fiber was normal, the morphology of the collagen fiber was normal, After 7d irradiation, the collagen was arranged in order and the density of collagen. The area of 28 d collagen fiber increased after IPL irradiation, and the difference was statistically significant( P<0.05).Conclusions(1) IPL exposure to the dermal layer of the rabbit skin has a certain stimulation, after irradiation 15 d can make the epidermis thick, after irradiation 28 d dermis has thickened.(2) IPL irradiation in rabbits after 7 days can make cuticular base cell layer of necrotic cells and dermal papilla layer and hair follicles surrounding hyperemia, edema,inflammatory cell infiltration, topical ketoprofen can effectively controlled inflammation after irradiation.(3) IPL wavelength of 620nm~1200nm irradiation of rabbit hair follicles at the same time,after irradiation 28 d collagen fiber increased.
