Objective:The effects of Biodentine on proliferation, osteogenesis of MG-63 cellsMethods The superficial area S=75mm2 of cylindrical research material is placed in a 1.5 ml culture extracting for 24 hours. After that,it is filtrated and sterilizated to make Biodentine extracts which concentration is 50 mm L / ml. 1:1 was continuously diluted for four times, and the dilution of five concentrations are: 1, 1/4, 1/8, 1/2 and 1/16. Biodentine were acted on MG-63 cells at different concentrations,then cell proliferation was detected by CCK-8 assay at 1, 3, 5 and 7 days. According to the results of cell proliferation experiments, 1/16 concentration of Biodentine was selected for MG-63 cells. The expression of osteogenic factor Runx2 m RNA in MG-63 cell was detected by Real-time PCR in 1,3,5 and 7 days.Then on the 10 th day and 14 th day after alizarin red staining, calcified nodules formed by MG-63 cells were observed.1.Effect of different Biodentine(BD) concentrations on the proliferation of MG-63 cells at different time points(1) Biodentine acts on MG-63 cells on day 1,the number of live cells compared with the blank control group, 1 concentration group, 1/2 concentration group and 1/8 concentration group decreased slightly; 1/4 concentration group has no obvious changes; 1/16 concentration group increased. There were no significant differences in the number of viable cells among the groups(p>0.05).(2) Biodentine acts on MG-63 cells on day 3,the number of live cells compared with the blank control group, 1 concentration group decreased slightly; 1/2 concentration group, 1/4 concentration group and 1/8 concentration group have no obvious changes; 1/16 concentration group increased. There were no significant diffterences in the number of viable cells among the groups(p>0.05).(3) Biodentine acts on MG-63 cells on day 5,the number of live cells compared with the blank control group, 1 concentration group decreased significantly, the difference was statistically significant(p<0.05); 1/2 concentration group decreased slightly; the difference was no statistically significant(p>0.05); 1/4 concentration group and 1/8 concentration group were slightly higher, the difference was not statistically significant(p>0.05); 1/16 concentration was significantly higher, the difference was statistically significant(p<0.05).(4) Biodentine acts on MG-63 cells on day 7,the number of live cells compared with the blank control group, 1 concentration group, 1/2 concentration group decreased significantly, the difference was statistically significant(p<0.05); 1/4 concentration group and 1/8 concentration group were slightly higher, the difference was statistically significant(p<0.05); 1/16 concentration group increased, the difference was not statistically significant(p>0.05). Results:(1) expression of Runx2 m RNA 1/16 concentration of BD was at 1st day of MG-63 cells, the expression of Runx2 m RNA in MG-63 cells was 1.14 times of the blank control group, and the difference was statistically significant(p<0.05); at 3rd day, the expression of m RNA Runx2 was 5.29 times of the blank control group, and the difference was statistically significant(p<0.05); at 5th day, the expression of m RNA Runx2 was 1.08 times of the blank control group, and the difference was statistically significant(p<0.05); at s7 th days, the expression of m RNA Runx2 was 2.11 times of the blank control group, and the difference was statistically significant(p<0.05).(2) Different groups of alizarin red staining 1/16 concentration of BD was at 10 th days of MG-63 cells, compared with the blank control group,no mineralized nodules were observed in the naked eye. Under the microscope, the small area red calcification in 2~3 can be seen. There was no significant difference between the two groups. 1/16 concentration of BD was at 14 th days of MG-63 cells, the blank control group showed that the large areas of red calcification in the 6 hole plate were found.; 1/16 concentration of BD group was red calcified, the area is wider and the color is deeper than the control group. 2.The effects of Biodentine on osteogenesis of MG-63 cellsConclusion: 1. Effect of different concentrations of Biodentine on cell proliferation in different. 50 mm2/ml Biodentine can inhibit cell proliferation,and the inhibition of cell proliferation was not significantly inhibited by the lower concentrations.. 2. Biodentine can increase the expression of m RNA Runx2 in MG-63 cells,and can strengthen the osteogenic effect of MG-63 cells.
