Objective: Gastric cancer is the most common malignant tumor of digestive tract. At present, methods for the treatment of gastric cancer are mainly surgery, radiation and chemotherapy, etc,which are far from satisfactory for the advanced gastric cancer. In recent years, targeted therapy based on specific molecular of malignant tumor has gradually become a hot research topic, and molecular target therapy for gastric cancer also causes more and more attention. The purpose of this report is to study the expression of EGFR and its mutant EGFRvⅢ protein in gastric cancer, and to analyze the relationship between their expression and clinical pathological features, aiming to probe into their roles in the occurrence and development of gastric cancer. It is hopeful to provide new targets and theoretical basis for the molecular target clinical treatment of gastric cancer in the near future.Methods: 95 cases of specimens of gastric cancer surgically resected and confirmed by pathology were collected.And we selected 47 cases with tissue adjacent to carcinoma as a contrast. EGFR、EGFRvⅢ protein expression in gastric cancer and cancer adjacent tissues have detected by immunohistochemistry. We analyze the relationship between the two kinds of protein expression and clinical pathological features by the chi-square test and bivariate Spearman correlation analysis of SPSS19.0 statistical software. Results with P < 0.05 said the difference was statistically significant.Clinical pathologic stage uses the P-TNM staging of the international cancer alliance revising in 2010.Results:1. The expression of EGFR protein in Gastric cancer tissue and tissue adjacent to carcinoma. The positive expression rate of EGFR in gastric cancer tissues and tissue adjacent to carcinoma was respectively 66.32%(63/95), 34.04%(16/47), the difference between the two groups was statistically significant(P<0.05). 2. The expression of EGFRvⅢ protein in Gastric cancer and tissue adjacent to carcinoma.The positive expression rate of EGFRvⅢ in 95 cases of gastric cancer tissues was 47.37%(45/95).However, we didn‘t detect the expression of EGFRvⅢ protein in tissue adjacent to carcinoma. The difference between the two groups was statistically significant(P<0.05). 3.The relationship between the expression of EGFR protein in gastric cancer and clinical pathological features.The positive expression rate of EGFR protein in male and female patients were respectively 67.14%(47/70) and 64.00%(16/25), the difference had no statistical significance(P > 0.05);In age ≤60 years or with age > 60 patients were respectively 63.33%(19/30), 67.69%(44/65), the difference had no statistical significance(P > 0.05);In high,medium differentiated and poorly differentiated groups were respectively 64.00%(16/25), 67.14%(47/70), the difference had no statistical significance(P > 0.05);In with lymph node metastasis group and without lymph node metastasis group were respectively 73.91%(51/69), 45.15%(12/26), lymph node metastasis group were significantly higher than those without lymph node metastasis, the difference was statistically significant(P < 0.05);In the invasion and serosa layer group and no invade and serous layer in the group were respectively 66.27%(55/83), 58.33%(7/12), the difference had no statistical significance(P > 0.05);In clinical pathologic staging Ⅰ-Ⅱperiod and stage Ⅲ-Ⅳ group were respectively 76.92%(10/13), 64.63%(53/82), the difference has no statistical significance(P > 0.05). 4.The relationship between the expression of EGFRvⅢ protein in gastric cancer and clinical pathological features.The positive expression rate of EGFRvⅢ in male and female patients were respectively 47.14%(33/70), 48%(12/25), the difference had no statistical significance(P > 0.05);In the age ≤60 patients and age > 60 years old were respectively 43.33%(13/30), 49.23%(32/65), the difference had no statistical significance(P > 0.05);In high, medium differentiated and poorly differentiated groups were respectively 28.00%(7/25), 54.29%(38/70), the difference was statistically significant(P < 0.05);With lymph node metastasis group and without lymph node metastasis group were respectively 55.07%(38/69), 26.92%(7/26), the difference was statistically significant(P < 0.05);In the clinical pathologic staging Ⅰ stage-Ⅱ and staging Ⅲ-Ⅳ were respectively 15.38%(2/13), 52.43%(43/82), the difference was statistically significant(P < 0.05);In the invasion and serosa layer group and no invade and serous layer group were respectively 51.80%(43/83), 16.67%(2/12), the difference was statistically significant(P < 0.05). 5.The relationship between the expression of EGFR and EGFRvⅢ protein in gastric cancer. The correlation coefficient of EGFR and EGFRv Ⅲ protein expression in gastric cancer tissue is 0.319(p = 0.002) with double variable Spearman correlation analysis, EGFR and EGFRv Ⅲ protein expression in gastric cancer tissue were positively correlated in the statis.Conclusion: 1. The expression of EGFR protein in gastric cancer tissues increased, the expression of EGFRvⅢ protein in gastric cancer tissues have positive,EGFR and EGFRvⅢ protein may have a certain relationship with the incidence of gastric cancer. 2.Gastric cancer expressed EGFR is easily to occur lymph node metastasis. 3. EGFRvⅢ protein may be involved in the differentiation, invasion and metastasis and has a certain relationship with prognosis of gastric cancer.
