Objective Obesity increases the incidence of many diseases. Breast cancer is one of the common malignant tumors in women. As a risk factor, obesity is related with breast cancer and plays an important role in the occurrence of breast cancer. But the mechanism of this effect remains unclear. Recent studies have demonstrated that leptin/OBR、PPARγ、 PI3K/Akt and other pathways are associated with obesity and breast cancer. Therefore, their roles in obesity related breast cancer deserves further study. The purpose of this study is to investigate the mechanism of leptin/OBR、PPARγ and PI3K/Akt in obesity-related breast cancer and provide reliable experimental and theoretical basis for the clinical prevention and treatment of breast cancer.Methods Specimens of breast cancer have been collected from hospital and were divided into two groups according to BMI (body mass index), BMI≤25 for the normal weight group and BMI> 25 for the obesity group. Immunohistochemical (SABC) was used to detect the protein level of PPAR y, Leptin, OBR, PI3K and p-Akt. RT-PCR method was used to detect the PI3K mRNA expression. Statistical analysis:statistical analysis was performed with SPSS, x2 test was used to evaluate the experimental results.Results1. Compared with normal weight group, the protein level of PPAR y in obesity group is decreased.2. There was a significant difference between the normal weight group and the obesity group in the expression of leptin. OBR was expressed in the cancerous tissue of the normal weight group and the obesity group, no significant difference was found between the two groups.3. The positive expression of PI3K and p-Akt protein in the obese group in the average optical density is significantly higher than the normal weight group.4. The results of RT-PCR show that the mRNA expression of PI3K is significantly elevated in cancerous tissues of the obesity group compared with the normal weight group.Conclusions1. The differential expression of PPARy and leptin in obesity group suggests that they might be involved in the obesity-related breast cancer.2. PI3K/Akt expression in the obesity group is higher than that in the normal weight group; speculate that this signaling pathway may play an important role in the occurrence and development of obesity-related breast cancer.
