Objective:To study the effect of Nutmeg extract on chronic cerebral hypoperfusion induced memory impairments.Methods:In the first experiment, chronic cerebral hypoperfusion was induced in male Wistar rats by bilateral common carotid artery occlusion(BCCAo). A Morris water maze was used to detect the impact of the changing of spatial learning and memory; Using immunohistochemical staining, observe the effect of nutmeg extract on pritein expression of mTOR; Using the ELISA, observe the effect of nutmeg extract on expression of NGF; The use of Western blotting techniques, to detect the changes of hippocampal ERK and pERK level.Results:1. Water maze test results showed that, compared with the control group, model group, the swimming distance was significantly prolonged, there was significant difference between the two groups (P< 0.01); compared with the model group, the treatment group was significantly shorter. The swimming distance, there was a significant difference between the two groups (P< 0.01), the ability of spatial learning and memory improved obviously.2. The results of ELISA show that compared with the control group the hippocampus region of rat nerve growth factor expression was significantly reduced; compared with the model group, the expression was significantly higher in rats hippocampus NGF area, there was a significant difference between the two groups (P< 0.05).3. The detection results of Immunohistochemical staining and western blotting, the examination of mTOR protein of Nutmeg extract could inhibit the chronic cerebral hypoperfusion induced.4. The western blotting examination revealed that Nutmeg extract could inhibit the protein expression of pERK induced by chronic cerebral hypoperfusion. There were significant differ-rentces between the model group and control group,between Nutmeg group and the model group.Conclusion:1. Nutmeg extract can effectively improve cerebral hypoperfusion deduced cognitive dysfunction and damage, can enhance the memory effect.2. Nutmeg extract can inhibit the cerebral hypoperfusion induced hippocampus nerve growth factor expression reduce, is conducive to the growth and development of neurons.3. Nutmeg ethanol extract could decrease the expression of mTOR protein could inhibit cerebral hypoperfusion induced brain tissue, inhibit neuronal necrosis and loss.4. Nutmeg ethanol extract can increase the level of inhibition of cerebral hypoperfusion induced phosphorylation of ERK in hippocampal tissue, showing its neuroprotective effect...
