Purpose:Researching protective effects of ginkgolide against intestinalischemia/reperfusion in rats brainMethods:120rats was divided into control group and solvent controlgroup, ginkgolide low dose group, ginkgolide dose group, the ginkgolidehigh dose group, ginkgolide3group was given internal carotid artery clip,bolt caused by carotid arterial line rat middle cerebral artery ischemia,ischemia after1h to pull out the middle cerebral artery in line, in order torealize the middle cerebral artery blood reperfusion, perfusion time for24hours, the control group (12rats) in addition to the middle cerebral artery isnot plugged, more than the rest of the steps were same as above, theinternal carotid artery in rat was cliped, model group was gaven line plugmethod, only given the same dose of0.9%normal saline within (excludingginkgolide),watching groups rats neurological symptoms at the same time,according to the Zea Longa five-point scale score method gave a mark onrats.After gaving a mark taked out the rats’ brain, placed in2%of dyesolution dyed GongSi azole nitrogen (TTC), infarction areas appear white,the infarction area red, measured range of infarction rats of whole brainpercentage (%) in rats. Extracted ischemia rats tissue homogenate,measured ginkgolide on brain tissue vitality of super oxide dismutase(SOD), malondialdehyde (MDA) content, and glutathione (GSH) content.Results:(1) ginkgolide impacted on the behavior of rats: controlgroup rats had no abnormal behavior, neurobehavioral score was gaven0points and solvent control group of rats appeared on the left side of the front PAWS to the outer circle, cannot be fully extended, or to the left sideof the dumping of nerve damage symptoms, behavior scores were1.7±0.6points, cerebral ischemia1h to injected ginkgolide sublingual vein of low,medium and high three dose group could significantly reduce ratsneurobehavioral scores, and dose, the higher the degree of group and lowerscore.(2) the ginkgolide effecting on cerebral infarction volumesize:control group rats had no infarction, solvent control group ratsischemic cerebral appearded infarction, about infarction volume is14.82%of the whole brain, high, medium and low three ginkgolide dose groups allshowed the ischemic infarction volume,Were reduced by10.72%,23.28%and29.49%respectively, and the higher dose of the more narrow lateralischemic infarction volume in rats, indicateed that ginkgolide onischemia/reperfusion injury in rats caused by infarction rats had asignificant effect.(3) ginkgolide on ischemic side of brain tissuehomogenate SOD vigor, MDA content, the influence of the content of GSH:determination of the content of MDA in group rats decreased, the vitality ofSOD and GSH content increased, suggests that the drugs on the body’sability to remove free radicals and antioxidant ability had improved.Conclusion: Many experiments had shown that ginkgolide wasplatelet activating factor (platelet activatingfactor, PAF) and calciumchannel blockers, it thus inhibited ischemia by inhibiting PAFfunction,ischemia produced the various abnormal inflammatory reactionand the formation of thrombosis. This study observed the ginkgolide oncerebral ischemia and protect function. Results showed that the ginkgolidecould reduce ischemia/reperfusion rats produced by neurological symptoms,reduced the scope of the cerebellar infarction increased lateral ischemictissue homogenate of rats the activity of SOD, increased the content ofGSH, reduced the MDA content, showed that ginkgolide could strengthen the ability to remove the body of free radicals, inhibited lipid peroxidationassociated, thus reduced the injury of brain tissue. it had obvious effect onbrain protection.
