Objective:Through detecting the change of IL-2、IL-4、CD3+、CD4+、CD8+of RhCantigen negative patients after infused RhC antigen positive blood, study on theeffects of RhC antigen to the immune function of recipients.Methods:We Collected622samples patients with emergency surgeries of the firsthospital of Jilin university from June2013to December2014, the range of agefrom21to40. Detect RhC antigen of patients before blood transfusion by themethod of micro column gel and salt solution media. Found the number of RhCantigen antigen negative samples is60. Over the same period in the bloodsupply of blood donors specimens in the center of the Changchun city, by themethod of micro column gel and saline medium method, two methods ofdetecting RhC antigen, choose the RhC antigen negative samples30,30RhCantigen positive samples as donors specimens. Research is divided into twogroups: one group is RhC incompatible group,30cases for the Rh system Cantigen negative recipients RhC antigen positive blood donors blood infusion,both for the RhC antigen incompatible transfusion. Another group is RhCcompatible group,30cases for RhC antigen negative blood transfusion Rh Cantigen negative blood donors, both for the RhC antigen compatible transfusion.We must also collect recipient samples form the RhC incompatible group and the RhC compatible group after24hours of transfusion. Using ELISA methodof IL-2and IL-4, using flow cytokines to detect CD3+, CD4+, CD8+cells.These immune function indicators were analyzed by statistical software,and thechange of immune function of these two group patients after type bloodtransfusion were compared.Results:1. IL-2in RhC incompatible group decreased24hours after the transfusion.10pg/ml was average reduce amount. IL-4in the group was increased8pg/mlaveragely. Compared with these indicator in blood samples before transfusion,there was statistically difference(p<0.05). IL-2in RhC compatible groupreduced1pg/ml averagely. IL-4in RhC compatible group increased1pg/mlaveragely. There was statistically difference before and after blood transfusion(p>0.05).2. CD3+in RhC incompatible group had the reduce ration13,with thereduce ration of CD4+is6and CD8+is3respectively.There was statisticallydifference before and after blood transfusion (p<0.05).Reduce ration of CD3+,CD4+, CD8+in RhC compatible group were3,2,2respectively and there was nostatistically difference before and after blood transfusion(p>0.05).3. Keep track of the study in the60of clinical blood transfusion cases, withcompatible group of30cases, in compatible group of30cases. They all had noclinical symptoms.Conclusion:1.The value of cytokines IL-2decreased and IL-4increased in the RhC antigen of the incompatible group after blood transfusion of24hours, causingthe immune function of the recipients have been suppressed.2. The value of cytokines of CD3+、CD4+、CD8+cells decreased, in theRhC antigen of the incompatible group after blood transfusion of24hours,causing the immune function of the recipients have been suppressed.3. The incompatible group make its receptors memory T cells form RhCantigen different transfusion.When the secondary blood transfusions havecaused effects on the immune response, may produce antibody of RhC, make theblood transfusion reaction.
