Pueraria flavonoids (PF) were the effective parts of kudzu root, as the continuousdevelopment of medical science, the pharmacological effects and health care functionof PF were gradually cognitive by people. The extraction, separation and purificationof PF were the key means for it’s utilization, to reduce the extraction process andimprove the extraction yields of PF, the study discussed the feasibility of extractingPF by high shear dispersion emulsifier with microemulsions as solvents; And insteadsynthetic surfactants by endogenous human bile acid salts to reduce the toxicity of alarge number of synthetic surfactants brought to the human body.High shear dispersion emulsifier was used to extract the active components ofkudzu root, the results showed that the extraction yields of puerarin, mirificin,3’-methoxyl-puerarin, daidzein and PF in70%ethanol were more than that ofmicroemulsion, while the daidzin showed the highest extraction yields inmicroemulsion. Central composite design-response surface method (CCD) wascarried out to select the optimal extraction screening:1:10~1:12.86(W/W) of solid-liquid ratio,0~11.43times dilution time of microemulsion, the most PF yields wereobtained at conditions of1:10of solid-liquid ratio,10times dilution time and24℃extraction temperature. The PF extraction yields by reflux method and ultrasonicmethod was much less than that of high shear dispersion emulsifier methods, whichwas234.51±2.7、237.31±3.35and (285.82±0.36) mg·g–1respectively.The excipients which had the most solubility of PF were selected, ternary phasediagrams and pseudo-ternary phase diagrams were constructed to optimizedformulations of microemulsion:linoleic acid glyceride:RH40:1,2-propyleneglycol=1:3:6(W/W/W). The synthetic surfactants in self-microemulsion werereplaced by endogenous human bile acid salts on the bases of optimized formulation.As the substitution ratio of bile acid salts increased, the clarify degree and light transmittance decreased, the pH of self-microemulsion containing NADC and NATCwere decreased gradually, while the pH of microemulsion which contained SCdisplayed an irregular change; the particle size of microemulsion comprised by SCwas increased slowly, while that microemulsion including NADC and NATCexhibited uneven alteration,the droplet size of microemulsio n was minimum with10.69nm The drug loading of microemulsion containing NADC, NATC and SCshowed jagged discrepancy, and achieved the maximum with21.99%when thereplacement ratio of when synthetic surfactants was swapped out with NATC at ratioof5:5(W/W). In conclusion, the various indicators of microemulsion were best whensynthetic surfactants was swapped out with NATC at ratio of5:5(W/W). The effect ofsodium deoxycholate (NADC), sodium taurocholate (NATC), sodium cholate (SC) onthe extraction yield of PF were investigated, the results revealed when70%ethanolcontaing cholates showed increased effect on the extraction yields, while that of10times diluted microemulsion showed decreased effect on the extraction yields of PF in10times diluted microemulsion were much more than that of70%ethanol at thesame concentration of cholates.The feasibility of extracting PF by high shear dispersion emulsifier withmicroemulsions as solvents was better, the extraction time could be shorten, at sametime increasing extraction yield. Compared with our previous research results,thedrug loading of self-microemulsion was improved from17.96%to21.99%afterreplaced by NATC,the content of synthesis emulsifier was reduced from45%to15%.
