Diabetes (diabetes, DM) is a kind of metabolic disease, as the maincharacteristics of insufficient insulin secretion and obstruction of utilization.The typical symptoms of diabetes is “Three mores and one less disease”, but underthe condition of continuous high glucolipid metabolic disorder, type Ⅱ diabetesoften involve multiple organs, such as eye and kidney, cardiovascular system, etc.Retinopathy is one of the most common complications for type Ⅱ diabetes, mainlybecause of the continuous hyperglycemia,hyperlipidemia. Currentlydiabeticretinopathy has become one of the main causes of blindness worldwideDiabetic retinopathy is one of the important in diabetes complications whichcan cause blindness disease. Firstly, the rise of blood glucose cause the change ofcrystal osmotic pressure and diopter leading to decreased visual and Blurring ofvision. If it is not well treated, proliferation of new blood vessels and fibrousproliferation will cause the retinal detachment, lead to permanent eye damage. Dueto its complicated pathogenesis, the consequence is usually very serious, which hasalready got wide attention in the world today. The main ingredients of QizhiJiangtang Capsule are radix astragali, radix rehmanniae, rhizoma polygonati, leech,which is mainly used in the treatment of Ⅱ diabetes and its complications. Studieshave confirmed that it has significant curative effect on diabetic nephropathy,but there is not any report about the effect on diabetic retinopathyThe experiment use Wistar rats with STZ to create the experimental diabeticrats’ models and use HUVEC with H2O2to create the experimental oxidativedamage models to study the related mechanism of Qizhi Jiangtang Capsule at themolecular level and provide new experimental basis for type II diabetes mellitus pathogenesis research and related disease prevention.In vivo part, the experiment useWistar rats with STZ to create the experimental diabetic rats’ models to create theexperimental oxidative damage. Wistar rats are randomly divided into normalcontrol group, diabetic model group,Qizhi Jiangtang Capsule high, medium and lowdose group, Shenqi Jiangtang Capsule is used as positive control group. Aftertreatment, lipid metabolism in rats needs to be detected. At the same time, the effectsof drugs on diabetic rats retina need to be detected by HE staining andimmunohistochemical technique. The quantity of protein expression as VEGF, PGI2,vWF are detected by ELISAIn vitro experiment, we use HUVEC with H2O2to createthe experimental oxidative damage models. Cellsare randomly divided into normalcontrol group, diabetic model group,Qizhi Jiangtang Capsule high, medium and lowdose group. After treat, using MTT to detect the cell survival rate; using FCM todetect the ROS; using ElISA to detect NO, NOS, SOD and VEGF; using RT-PCR todetect the mRNA level of Bcl-2, ET-1, eNOS. The results show that thelipid metabolism in the groups of Qizhi Jiangtang Capsule high, medium and lowdose, Shenqi Jiangtang Capsule positive control are significantly improved. VEGFand AVG-2in Qizhi Jiangtang Capsule high, medium and low dose byimmunohistochemical technique are significantly decreased compared with normalgroup. But diabetic rats retinadetected by HE staining have no significant changes;the expression of PGI2is higher and VEGF and vWF are lower in Qizhi JiangtangCapsule high, medium and low dose, Shenqi Jiangtang Capsule positive controlgroup than model group.In vitro experiment shows that compared with model group,the groups of Qizhi Jiangtang Capsule high, medium and low dose can improve theoxidative damage modelsmade by H2O2, reduce ROS concentration, increase theexpression of NO, NOS and SOD in cells, up-regulate the mRNA expression ofBcl-2and eNOS, and down-regulateET-1and NOX-4at the same time.In conclusion,studies confirm that Qizhi Jiangtang Capsule can improve the type Ⅱ diabetes lipidmetabolism by improving retinal vascular endothelial and inhibit retinalneovascularization formed in order to achieve a therapeutic goal...
