Hoya carnosa, a common traditional medicinal material, is widely distributed in Fujian area, which has unique curative effect to a variety of diseases. It has wide applications. In order to effectively develop and use this plant resource, we try to do the following experiments:1Effect of antibacterial activity on Hoya carnosaWe get5extracts from Hoya carnosa, they are stem water extracts, stem80%ethanol extracts, leaf water extracts, leaf80%ethanol extracts and leaf juice extracts. We test their antibacterial activity. Result:Stem80%ethanol extracts have better inhibition effect in streptococcus and Aeromonashydrophila, stem water extracts have better inhibition effect in Aeromonashydrophila, leaf80%ethanol extracts have better inhibition effect in Aeromonas haianesis, sp. Nov, leaf water extracts have better inhibition effect in Escherichia coli, leaf juice extracts have better inhibition effect in Aeromonas trota. In a word, all these extracts have antibacterial activity to some extent.2Verify chemical materials in those extractsWe use routine chemical methods to verify chemical materials in above5extracts. Result:Stem80%ethanol extracts may contain organic acids, saponins, polysaccharides, flavonoids, coumarins, steroids, etc. Stem water extracts may contain organic acids, saponins, amino acid, polypeptide, polysaccharides, coumarins, steroids, etc. Leaf80%ethanol extracts may contain organic acids, polysaccharides, flavonoids, coumarins, steroids, etc. Leaf water extracts may contain organic acids, saponins, amino acid, polypeptide, steroids, etc. Leaf juice extracts may contain organic acids, amino acid, polypeptide, polysaccharides, flavonoids, cardiac glycosides, coumarins, steroids, etc. In a word, the extracts mainly have saponins and organic acids.3Extraction and purification of saponins on Hoya carnosaWe get saponin using traditional extracting methods, after bubble test and color showing test, we proof it as saponin. Evident, using this method to separate saponin is feasible. 4Pharmacological actions of saponins on Hoya carnosaWe try to test vitro antioxidant and hemolysis effect. Result:The saponin has total vitro antioxidant ability, it also can clear O2-·free radicals, max rate is63.20%. The saponin also has hemolysis effect to some extent, it can cause damage to blood cells, minimum concentration is2mg/mL. Sapomns have vitro antioxidant and outside body hemolysis effect.5Study physiology and biochemistry effect of saponin on miceWe test antitussive and expectorant effect on mice, whose cough is causing by ammonia water. We also test their body antioxidant effect and serum biochemical indexes. Result:Saponin could prolong cough incubation period and reduce cough frequency in mice within3mins, also increase volume of trachea fluid, obviously enhance SOD, GSH-Px ability, obviously decrease the content of MDA, significantly increase content of total protein and AKP, also significantly reduce the content of urea nitrogen. All in all, the saponins have antitussive and expectorant effect to some extent, can enhance body antioxidant effect available.6Study condition of purifying saponin in Hoya carnosaWe use PHPLC to find out the conditions of extracting and purifying saponin. We get the preliminary PHPLC conditions:PHPLC column:pursuit XRs C18, mobile phase:acetonitrile-water(14:86), column temperature:almost20℃, flow:1mL/min, detection wavelength:203nm and210nm.This experiment is trying to find out the ability that saponins has, in order to offer reference in promoting clinical practice of Hoya carnosa.
