| Acanthopanax sentious is one of the traditional Chinese medicines, which has very important research value. Contemporary medical researches prove that Acanthopanax senticosus has a variety of pharmacological effects as a result of saponins which is the major components there are many biological activities, such as antibacterial, antiviral and so on. It has been used in household chemicals, pesticides and food industry widely. So it has a huge advantage of prospect. In this paper, Acanthopanax sentious leaves are raw materials.To get low sugar chain, highly activity of Acanthopanax saponins for the purpose. The extraction and purification technology of Acanthopanax sentious saponins are researched. The strains are sifted from Aspergillus, which enzyme solutions were able to hydrolyze Acanthopanax sentious saponins. The best enzyme solutions conditions were determined. Acanthopanax senticosus saponins were separated by Silica gel column. The new saponins were obtained by silica of1mm and they were analyzed. Specific results are as follows:The influence factors on the extraction of Acanthopanax senticosus saponins were examined. The factors include extraction temperature, soaking time of ethanol, ethanol volume fraction and ratio of material to liquid (g/mL). Acanthopanax senticosus saponins were purified through the method of column chromatography, the volume flow rate and ethanol elution volume fraction was optimized. In the result, the optimal extracting conditions were obtained:extraction temperature was80℃, soaking time was5h, ethanol volume fraction was70%, and ratio of material to liquid was1:6. The best conditions for purification were ethanol flow rate2.72×10-5m3/min and ethanol elution volume fraction60%. At the optimal conditions of extraction and purification, the yield and the content of Acanthopanax senticosus saponins were5.9%and44.1%, respectively.In order to screen the strains which can produce the hydrolase of Acanthopanax saponins, ten kinds of microbial strains were studied in this paper. And the optimal condition of enzyme hydrolysis was also studied through orthogonal design with four factors and three levels. The saponins were analyzed by TLC. Aspergillus niger UV-11s and Aspergillus oryzae sp.39s were screened as the stains which can hydrolyze by Acanthopanax saponins.The optimal conditions of Aspergillus niger UV-11s that enzyme Acanthopanax senticosus saponins, reaction time was40h, reaction temperature was30℃, the concentration of saponins was3.0mg/mL, and pH value was6.0; The optimal conditions of Aspergillus oryzae sp.39s those enzyme Acanthopanax senticosus saponins:reaction time was24h, reaction temperature was50℃, the concentration of saponins was3.5mg/mL, and pH value was6.0.The hydrolyzed Acanthopanax senticosus saponins were separated in Silica gel column. The new saponins were obtained by silica of1mm, after analysis by TLC, a new saponins that Rf=0.77,0.50was obtained, and the structure of the new saponins needs analysis farther more. |
