Objective: To study the expression of Annexin II in different status of dermalpapillae cells in vitro.Methods: Dermal papillae cells was isolated from the scalp specimen of healthypeople after facial plastics surgery by two-step enzyme digestion and cultured in vitro. Thecells were classified into low-passage dermal papillae cells with aggregation andhigh-passage dermal papillae cells with nonaggregation according to the number ofpassage and growth characters. Cellular TRNA was isolated by Trizol method and appliedfor RT-PCR to detect the expression of Annexin II at mRNA level. Moreover, theexpression of Annexin II at protein level was examined by Western-Blot.Results:(1) Compared with that in high-passage dermal papillae cells withnonaggregation, the expression of Annexin II in low-passage dermal papillae cells withaggregation was significantly higher.(2) At the protein level, Annexin II has2differentisoforms, Annexin II isoform was found in both high-passage dermal papillae cells withnonaggregation and low-passage dermal papillae cells with aggregation, while AnnexinII was only found in low-passage dermal papillae cells with aggregation.Conclusions: The expression of Annexin II was possibily inhibited in high-passagedermal papillae cells with nonaggregation by post-transcription mechanism which needs tobe further confirmed. Its expression in low-passage dermal papillae cells with aggregationmay play a role in aggregation growth of low-passage dermal papillae cells.
